Abstract Strawberry shoot meristem cultures have been used for producing virus-free strawberry plants. Plantlet regneration from leaf mesophyll protoplasts, and Agrobacterium tumefaciens mediated genetic transformation, mark the beginning of strawberry improvement with biotechnologies. The use of biotechnological tools has a greater potential in producing strawberry plants with traits such as early maturation, frost and disease resistance, mite and nematode resistance, high yield, better quality, suitability for mechanical harvesting, shipping ability, ellgaic acid content, aroma compounds and cost-effective micropropagation. Gene identification and mapping with RFLPs or RAPDs would be desirable. Transgenic strawberries should be tested for their quality before releasing to the consumers. 相似文献

Cottonhead windhairdaisy (Saussurea laniceps Hand.-Mazz.) is one of the most famous and important medicinal herbs in China. Illegal collection from wild populations is increasingly threatening the present environment of S. laniceps. Establishment of an efficient method for micropropagation is the best way to change its endangered situation. When mature seeds of S. laniceps were cultured on hormone-free MS medium, plantlets were formed from germinated seeds in 7–10 d. Then 0.5 cm × 0.5 cm leaf explants were transplanted to MS medium supplemented with 1-naphthalene-acetic acid (NAA)/2,4-D and benzyladenine (BA)/KT and callus was achieved 10 d after transfer. Shoot bud regeneration occurred from callus cultured on MS medium supplemented with different growth regulators 20 d after culturing. The regeneration percentages varied with the different components of plant growth regulators. The percent regeneration from callus pretreated at low temperature of 5°C increased significantly compared with those incubated at 23/20°C directly. Optimal regeneration was observed with explants on media supplemented with 1.5 mg8226;L–1 BA plus 0.2 mg8226;L-1 NAA. In the presence of 0.2 mg8226;L–1 NAA in half-strength MS, 78% of the shoots formed roots. Plantlets from explants showed 63% survival after acclimatization. 相似文献

Black locust (Robinia pseudoacacia L.) is one of the most important stand-forming tree species in Hungary and its importance is increasing in many countries. The main aim of the discussed new selection programme is to identify black locust clones with good performance and good form for setting up clonal seed orchards. As a result of selection programme 16 new black locust clones have been improved. In spring 2002 a black locust seed orchard was established with the newly selected clones. About 40% of the plants can be considered to belong to the height growth rate class 1 and 2. Four years' old micropropagated plants regenerated by enhanced axillary branching from shoot buds of a single genotype of
Robinia pseudoacacia were characterized by RAPDs. Random amplified polymorphic DNA analysis was carried outusing 19 random 10-mer DNA primers
and 286 RAPD bands were examined which showed 30% polymorphism. Similarity indices ranged from 0.86 to 0.96 among different
plants based on RAPD data. The UPGMA dendrogram was constructed based on similarity indices which showed clustering of different
plants into subgroups based on similarity values. Our results suggest that somaclonal DNA sequence variations are present
even when organized cultures such as shoot buds were used as explant for micro-propagation.

为实现组培微环境CO2 增施的长期自动控制 ,解决增施CO2 过程中可能将空气中杂菌引入组培箱的问题 ,设计开发了一套基于PLC的无糖组培微环境控制系统。系统以PLC为控制核心 ,采用定量供给的CO2 增施控制策略 ,可根据设定要求将固定量的高压、高纯度CO2 气体直接供入组培箱 ,并可对组培箱内湿度进行调控。系统调试试验结果表明 ,控制系统能将光期组培箱内CO2 摩尔分数和相对湿度分别控制在 70 0~ 90 0 μmol·mol-1和80 %~ 92 %的目标范围内 ,达到了预期效果 相似文献
