AIM:To investigate the potential role of p62dok in the regulation of hepatic gluconeogenesis. METHODS:The expression of p62dok, insulin signaling transduction, and hepatic gluconeogenesis were investigated in the liver tissues of mice treated with high-fat diet(HFD) and in cultured mouse hepatocytes treated with free fatty acid(FFA). The experiments of gene silencing and overexpression were conducted to observe the effects of p62dok on insulin signal transduction and hepatic gluconeogenesis in cultured mouse hepatocytes. Western blotting was used to detect the protein levels and the phosphorylation statue. RESULTS:The increased p62dok levels were found in the liver tissues from HFD-treated mice and FFA-treated hepatocytes. Meanwhile, phosphorylation of Akt and forkhead box O1 protein(FoxO1) was were decreased and the expression of glucose-6-phosphatase(G6Pase) and phosphoenolpyruvate carboxykinase(PEPCK) was increased. Silencing of p62dok in cultured hepatocytes treated with FFA induced the increase in phosphorylation of Akt and FoxO1, and decrease in the protein levels of G6Pase and PEPCK. CONCLUSION:Up-regulation of p62dok induced by HFD or FFA enhances hepatic gluconeogenesis via inhibiting insulin signal transduction.  相似文献

Blackgram fiber (Phaseolus mungo): Mechanism of hypoglycemic action   总被引:2,自引:0,他引:2  
The effect of the administration of blackgram fiber (Phaseolusmungo) on the metabolism of carbohydrates was studied in rats fed 30%NDF (neutral detergent fiber) diet. The experimental group showed a significant increase in liverglycogen level and a significant decrease in blood glucose. Significantincreases in the activities of glycogen phosphorylase, hexokinase, fructose-1,6-diphosphatase, glucose-6-phosphate dehydrogenase and isocitratedehydrogenase were observed in the experimental group. The activities ofphosphoglucomutase and glucose-6-phosphatase were significantly lower inrats fed the fiber diet. The study showed that blackgram fiber exhibitssignificant hypoglycemic action in experimental animals.  相似文献

The aim of this study was to elucidate the mechanisms underlying the glucose lowering effects of thymoquinone in streptozotocin (STZ)-induced diabetic hamsters in terms of hepatic glucose production. Diabetes was induced by intraperitoneal injection of 65 mg/kg body weight of STZ. Treatment with thymoquinone commenced 4 weeks after induction of diabetes at a daily dose of 50 mg/kg body weight by gastric gauge. Blood glucose and glycated hemoglobin levels were significantly reduced depending on periods of the treatment. Thirty days after commencement of thymoquinone-treatment, hepatocytes were isolated using collagenase to determine liver glucose production. Glucose production after 2 h incubation of the isolated hepatocytes with gluconeogenic precursors (alanine, glycerol and lactate) was significantly lower in hamsters treated with thymoquinone as compared to that in vehicle controls. The results of this study demonstrate that the antidiabetic action of thymoquinone is at least partially mediated through a decrease in hepatic gluconeogenesis.  相似文献

Summary

Following EDS'76 virus (BC14 virus) infection of breeder chickens by the conjunctival route, vertical transmission occurred in the first week after infection. In the progeny which had been infected with EDS'76 virus by the vertical route, increasing haemagglutination inhibiting (HI) litres to BC14 virus and increasing numbers of birds with HI litres were observed from 3 weeks to 15 weeks of age. Sixty‐one per cent of the hens and 77 per cent of the cocks had 2log HI BC14 virus litres exceeding 4 at an age of 15 weeks.

Some birds which had been serologically negative throughout the rearing period, seroconverted between 25 and 28 weeks of age. This phenomenon occurred in hens as well as in cocks. Simulation of stress twice during the laying period by injection of corticosteroid hormone did not increase the number of birds serologically positive to EDS'76 virus.

EDS'76 was observed in the group of hens that was vertically infected, since egg production was significantly depressed between 28 and 34 weeks of age. Probably this was mainly the result of a production drop in the hens showing seroconversion at 27 or 28 weeks of age.

In the group of fowl vertically infected with EDS'76 virus, serologically positive birds appeared to be protected for the greater part to BC14 virus challenge at 50 weeks of age, while negative birds seemed to be fully susceptible. Chicks hatched from eggs collected in the third and fourth week after infection of the dams had maternal antibodies. Fertility and hatchability of apparently normally shelled eggs seemed not to be affected after BC14 virus infection of the dams. Intensive contact with contaminated faeces is probably an indispensable condition for lateral transmission of the virus.  相似文献

The aim of this study was to elucidate the mechanisms underlying the hypoglycaemic effect of N. sativa oil (Nigella sativa oil) in streptozotocin (STZ)-induced diabetic hamsters, in terms of hepatic glucose production, and to investigate the possible immunopotentiating effect of N. sativa oil on peritoneal macrophages. Diabetes was induced by intraperitoneal injection of 65 mg/kg body weight of STZ. Treatment with N. sativa oil commenced 6 weeks after induction of diabetes at a dose of 400 mg/kg body weight by gastric gavage. Isolated hepatocytes were collected using collagenase to determine liver glucose production. Phagocytic activity was evaluated by injection of fluorescent latex (2 microm diameter) intraperitoneally, followed 24 h later by collection of peritoneal macrophages. N. sativa oil reduced blood glucose from 391+/-3.0 mg/dl before treatment to 325+/-4.7, 246+/-5.9, 208+/-2.5 and 179+/-3.1 mg/dl after the first, second, third and fourth weeks of treatment, respectively. Hepatic glucose production from gluconeogenic precursors (alanine, glycerol and lactate) was significantly lower in treated hamsters. Treatment with N. sativa oil significantly increased the phagocytic activity and phagocytic index of peritoneal macrophages and lymphocyte count in peripheral blood compared with untreated diabetic hamsters. Our data indicate that the hypoglycaemic effect of N. sativa oil is due to, at least in part, a decrease in hepatic gluconeogenesis, and that the immunopotentiating effect of N. sativa oil is mediated through stimulation of macrophage phagocytic activity either directly or via activation of lymphocytes.  相似文献

Nutritional regulation of hepatic glucose metabolism in fish   总被引:2,自引:0,他引:2  
Glucose plays a key role as energy source in the majority of mammals, but its importance in fish appears limited. Until now, the physiological basis for such apparent glucose intolerance in fish has not been fully understood. A distinct regulation of hepatic glucose utilization (glycolysis) and production (gluconeogenesis) may be advanced to explain the relative inability of fish to efficiently utilize dietary glucose. We summarize here information regarding the nutritional regulation of key enzymes involved in glycolysis (hexokinases, 6-phosphofructo-1-kinase and pyruvate kinase) and gluconeogenesis (phosphoenolpyruvate carboxykinase, fructose-1,6-bisphosphatase and glucose-6-phosphatase) pathways as well as that of the bifunctional enzyme 6-phosphofructo-2-kinase/fructose-2,6-bisphosphatase. The effect of dietary carbohydrate level and source on the activities and gene expression of the mentioned key enzymes is also discussed. Overall, data strongly suggest that the liver of most fish species is apparently capable of regulating glucose storage. The persistent high level of endogenous glucose production independent of carbohydrate intake level may lead to a putative competition between exogenous (dietary) glucose and endogenous glucose as the source of energy, which may explain the poor dietary carbohydrate utilization in fish.  相似文献

Juvenile gilthead sea bream were fed to visual satiety with isonitrogenous diets based on fish meal and different plant ingredients (33–35% replacement) supplemented with free amino acids to meet the desired indispensable amino acid (IAA) profile and dispensable amino acid (DAA) content. In diets M and WB, IAA profile and DAA content resemble that of the muscle or whole body, respectively. In diets MGlu and WBGlu, DAA content was increased by adding -glutamic acid (Glu) and thus the IAA/DAA ratio varied from 1.13 (diet M) to 0.80 (diet WBGlu). Growth rates were not significantly different among experimental groups, but feed conversion ratio and nitrogen retention were impaired by the decrease of dietary IAA/DAA ratio. Postprandial ammonia excretion increased with the increase of dietary DAA content irrespective of IAA profile. Conversely, hepatic activity of glutamate dehydrogenase (GDH) was lower in fish fed diet WBGlu than in fish fed diet M. Hepatic growth hormone (GH) binding was not significantly affected by the dietary treatment, but circulating levels of insulin-like growth factor-I (IGF-I) and GH were, respectively, down- and up-regulated in fish fed diet WBGlu, which suggests some defect in the transmission of GH receptor signal. Fat retention and hepatic activities of lipogenic enzymes (glucose-6-phosphate dehydrogenase, G6PD; malic enzyme, ME) were decreased in fish fed diet MGlu. Key metabolic enzymes of hepatic glycolysis (glucokinase, GK) and gluconeogenesis (phosphoenolpyruvate carboxykinase, PEPCK) were also altered in this group of fish. Since soybean meal concentration was highest in diet MGlu, results on lipid and carbohydrate metabolism can be primarily attributed to this component of the diet. In contrast, data on growth performance, ammonia excretion and GH axis mainly reflect changes in the dietary amino acid profile, which reveals that a muscle IAA profile and a high IAA/DAA ratio are important in feeds for gilthead sea bream.  相似文献
