AIM: To investigate the changes of myocardium glycogen content and the relation ship between changes of the myocardial glycogen content and the myocardial function. METHODS: Male Wistar rats were placed in (hypoxia rats) and made to swim in (hypoxia swimming rats) a hypobaxic chamber simulating an altitude of 5 000 m above sea level. The content of myocardium glycogens was determined by colorimetry. RESULTS: The myocardium
glycogen content of rats significantly reduced along with the prolongation of hypoxic exposure and approached to control level in hypoxia swimming rats. The myocardial function of right ventricule was improved significantly compared with control group.CONCLUSION: Moderate exercise (swimming) is beneficial to hypoxic adaptation of rats under the condition of chronic hypoxia.  相似文献

Several effects of hypertrehalosemic hormone (Peram-HTH) have been compared with the action of 1,2,3,4,5,6-hexachlorocyclohexane (γ-HCH) in the cockroach, Periplaneta americana. The data show that both agents stimulate respiration while maintaining an R.Q. of approximately 1. The action of γ-HCH differs from that of Peram-HTH-I in that it promotes oxidation of large amounts of soluble carbohydrate. Recent studies show that Peram-HTH-I has an equally potent stimulatory effect on lipid deposition in the fat body and on the release of trehalose from the same tissue. It is of interest that γ-HCH not only increases triacylglycerol accumulation in the fatγ body but that this effect is more pronounced if the γ-HCH is applied to the ventral surface of the mesothorax. γ-HCH also depleted whole body glycoprotein and glycolipid whereas Peram-HTH-I had no effect on glycoprotein but increased that of glycolipid. It is clear that γ-HCH causes complex disruptions to the metabolism of the insect, many of which are related to the release of hormones. The study supports the idea that certain actions of γ-HCH are mediated through the release of Peram-HTH-I.  相似文献

In an attempt to identify appropriate diets for use in intensive baitfish culture, the efficacy of three commercial diet formulations, a high-protein, high-lipid, low-carbohydrate diet [a semi-moist salmonid diet (SD)] and two low-protein, low-lipid, high-carbohydrate diets [a catfish diet (CD) and a tilapia diet (TD)] on growth performance of juvenile white suckers, Catostomus commersoni, was examined. All three diets permitted growth during the 16 week trial. The SD promoted highest growth and somatic protein and lipid deposition, whereas the CD and TD formulations promoted markedly higher accumulations of glycogen in the carcass and liver. The high hepatic glycogen content was associated with an elevated hepatosomatic index in the CD- and TD-fed groups. Plasma T3 levels were lower in the CD- and TD-fed groups, possibly a response to the low dietary protein or indicative of dietary insufficiency in these fish. Of the three diets evaluated, the SD was considered to be the best in terms of the growth performance parameters measured, and offers an economical and appropriate diet for intensive culture of juvenile white suckers.  相似文献

AIM:To investigate the effects of siRNA targeting integrin-linked kinase (ILK) on the expression of glycogen synthase kinase 3β (GSK-3β) and β-catenin during epithelial-mesenchymal transition (EMT) in human kidney proximal tubular epithelial cell line HKC induced by high glucose. METHODS:HKC cells were divided into 4 groups:normal glucose (NG) group, high glucose (HG) group, HG+HK (a vector containing the non-specific siRNA designed as negative control) group and HG+ILK siRNA group. The inverted fluorescence microscope was used to examine the expression of green fluorescent protein (GFP). The expression of ILK at mRNA and protein levels was detected by RT-PCR and Western blotting. The expression of p-GSK-3β and β-catenin was observed by immunocytochemical staining. The protein expression of total GSK-3β, p-GSK-3β, nuclear β-catenin, total β-catenin, E-cadherin and α-smooth muscle actin (α-SMA) was measured by Western blotting. RESULTS:GFP was observed in HKC cells, indicating that the transfection was successful. Both the protein and mRNA of ILK were down-regulated in HG+ILK siRNA group compared with HG group and HG+HK group, but still higher than those in NG group. Silencing of ILK down-regulated the expression of p-GSK-3β and nuclear β-catenin. No difference of total GSK-3β or total β-catenin was observed among the 4 groups. CONCLUSION:These data support a functional role of ILK, GSK-3β and β-catenin in tubular EMT induced by high glucose. ILK may promote tubular EMT by regulating the activity of GSK-3β and β-catenin, the downstream effectors of the Wnt/β-catenin pathway.  相似文献

AIM: To study the effect of chronic corticosterone (CORT) injection on the depression-like behaviors and the brain glycogen level in mice. METHODS: Male C57BL/6N mice (n=40) were randomly divided into normal control group and model group. The mice in model group were subcutaneously consecutively injected with CORT for 4 weeks. The mouse model of chronic stress depression was constructed. The forced swim test and open field experiment were conducted to prove chronic stress model. The serum level of CORT in the mice was measured by radioimmunoassay. The protein levels of hippocampal synaptophysin (SYP) and brain-derived neurotrophic factor (BDNF) were detected by Western blot. Hippocampus glycogen, glycogen synthase and glycogen phosphorylase were determined by indirect fluorescence measurement. RESULTS: Compared with normal control group, the immobility time of the forced swim test in model group was significantly lengthened (P<0.01), and the ability of spontaneous activity was reduced (P<0.01), indicating that chronic CORT injection induced depression-like behaviors in mice. The CORT level increased significantly (P<0.01) in model group. CORT injection decreased the protein expression of hippocampal SYP and BDNF (P<0.01), reduced hippocampal glycogen level (P<0.05) and glycogen synthase activity (P<0.05), and increased glycogen phosphorylase activity (P<0.05). CONCLUSION: Chronic CORT injection causes hippocampal neuron damage and induces the depression-like behaviors of mice, which may be associated with decreasing hippocampal glycogen level by CORT.  相似文献

[目的]为了鉴定马铃薯反义SSU的功能。[方法]利用分子生物学技术,构建了马铃薯反义SSUcDNA原核表达载体(pE-aS),通过0.1mol/LIPTG诱导pE-aS在大肠杆菌BL21(DE3)中表达,测定糖原的相对含量。[结果]经连接、转化和酶切鉴定筛选出重组表达载体pE-aS,而pE-aS转化BL21(DE3)获得重组菌株BL21-aS。经IPTG诱导的BL21-aS糖原含量OD值为0.409,显著低于对照菌株BL21(DE3)和未经IPTG诱导的BL21-aS,而对照菌株BL21(DE3)和未经IPTG诱导的BL21-aS的糖原含量OD值差异不显著。这表明反义SSU在BL21内表达能够抑制glgC编码的AGPase的形成,从而减少其糖原合成量。[结论]该研究为构建马铃薯反义SSU植物转化载体奠定了基础。  相似文献

The effect of dietary starch and fat content on serum creatine kinase (CK) activity and substrate availability was evaluated in 4 mares of Quarter Horse-related breeds with polysaccharide storage myopathy (PSSM). Four isocaloric diets ranging in digestible energy (DE) from 21.2% (diet A), 14.8% (B), 8.4% (C), to 3.9% (D) for starch, and 7.2% DE (diet A), 9.9% (B), to 12.7% DE (diet C and D) for fat were fed for 6-week periods (4 weeks with exercise) using a 4 X 4 Latin square design. Postprandial glucose and insulin responses were measured, and 4 hours postexercise, serum CK activity, glucose, insulin, free fatty acids (FFA), and beta-hydroxybutyrate (beta-HBA) were analyzed. Glycogen, glucose-6-phosphate, citrate synthase, 3-hydroxy-acyl-CoA dehydrogenase, lactate dehydrogenase as well as abnormal polysaccharide and lipid content were measured in middle gluteal muscle samples. Postprandial insulin and glucose response was higher for diet A versus D. Log CK activity was higher with diets A, B, and C versus D. Daily insulin was higher and FFA lower on diet A versus B, C, and D, whereas glucose varied only slightly with diet. Muscle oxidative capacity and lipid stores were low in PSSM horses and muscle glycogen and abnormal polysaccharide content high on both diets A and D. Individual variation occurred in the response of PSSM horses to diets differing in starch and fat content. However, for those horses with clinical manifestations of PSSM, a diet with <5% DE starch and >12% DE fat can reduce exertional rhabdomyolysis, potentially by increasing availability of FFA for muscle metabolism.  相似文献

AIM: To examine the effects of high glucose (HG) on the expression of Snail1 and protein kinase B (Akt)/glycogen synthase kinase 3β (GSK-3β) in primary renal tubular epithelial cells (RTECs). METHODS: The primary RTECs were randomly treated with normal glucose, high glucose or D-mannitol for 30 min~72 h. RT-PCR and Western blotting were used to observe the expression of Snail1, Akt and GSK-3β at mRNA and protein levels in these cells. The primary cultured RTECs were pretreated with LY294002 (a PI3K inhibitor, 25 μmol/L) to observe the specific inhibitory effects of phosphatidylinositol 3-kinase (PI3K) on HG-induced expression of Snail1 protein. RESULTS: Treatment of RTECs with HG resulted in increased mRNA and protein levels of Snail1, Akt1, and phosphorylation of Akt and GSK-3β. LY294002 blocked the HG-induced up-regulation of p-Akt, p-GSK-3β and Snail1 expression at protein level, but no effect of LY294002 was seen on the total protein expression of Akt1 and GSK-3β. HG did not affect the expression of GSK-3β at mRNA and protein levels. CONCLUSION: HG-induced up-regulation of Snail1 may be regulated by Akt/GSK-3β pathway in RTECs.  相似文献

AIM: To study the changes and correlations of β-catenin, multidrug resistance-associated protein 2 (MRP2), P-glycoprotein (P-gp), Bcl-2 and the free ATP level in esophageal squamous-cell carcinoma (ESCC) cell line EC-109 induced by ATP-competitive glycogen synthase kinase(GSK-3) inhibitor 6-bromoindirubin-3'-oxime (BIO). METHODS: The methods of flow cytometry, immunofluorescence, cytochemistry and ATP assay were used to study the expression levels of MRP2, P-gp, β-catenin and Bcl-2, the efflux capability of ATP-binding cassette (ABC) transporters, and the free ATP level in ESCC cells. RESULTS: After induced by BIO, up-regulation of β-catenin and Bcl-2 expression in cytoplasm and accumulation of β-catenin in nucleus were found in ESCC cells. The expression of MRP2 was up-regulated in cytoplasm and cell membrane. On the contrary, the expression of P-gp was down-regulated in cytoplasm and cell membrane. The free ATP level and the efflux capability of ABC transporters increased in ESCC cells. CONCLUSION: The multidrug resistance of ESCC cells in enhanced by BIO treatment.  相似文献

The aim of this study was to determine the prevalence of equine polysaccharide storage myopathy (EPSM) in two populations of horses in the UK. Biopsy specimens from 94 horses presented to an abattoir (population 1), and 46 horses with neuromuscular disorders presented to a university referral hospital (population 2) were obtained over a period of 4 years. Histological sections were examined by a veterinary pathologist for lesions including abnormal polysaccharide inclusions in myofibres.In population 1, a diagnosis of EPSM was made in 8% and non-specific myopathy in 33% of horses. In population 2, a diagnosis of EPSM was made in 22%, equine motor neurone disease (EMND) in 15% and non-specific myopathy in 37%. Within each population there was no difference in age, sex or breed distribution and muscle disease diagnosis. However, populations differed from each other in age and breed distributions and muscle disease diagnosis. EPSM was found in draft, Warmblood and related breeds and was diagnosed for the first time in cob-types. EMND was reported in 7/46 horses presented for neuromuscular disease and weakness, representing an important diagnosis in the UK. This study showed a high prevalence of EPSM and other myopathies in typical breeds of horses in the UK.  相似文献
