The effects of four cryoprotectants (methanol, MeOH; dimethyl sulphoxide, DMSO; dimethyl acetamide, DMA; and ethylene glycol, EG), three extenders (calcium‐free Hanks' balanced salt solution, C‐F HBSS, Hanks' balanced salt solution, HBSS and sodium chloride, NaCl) and two different freezing procedures (one‐ and two‐step) on the cryopreservation of striped catfish (Pangasius hypophthalmus (Sauvage)) sperm were investigated. Sperm were frozen using a controlled‐rate freezer in 250 μL straws and stored for 2 weeks in a liquid nitrogen (LN2) container. They were then airthawed at room temperature, and fertilization, motility and viability were assessed. The highest fertilization rate of 41% (81% of control) was achieved with the combination of 12% DMSO and 0.9% NaCl using a one‐step freezing procedure (10°C min?1). Also, DMA resulted in a higher fertilization rate (30% or 51% of the control) than MeOH (18% or 38% of the control) or EG (8% or 12% of the control). In addition, the three extenders used did not affect fertilization rates after cryopreservation with each cryoprotectant. There were no significant differences among the three cryoprotectant concentrations and between the one‐ and two‐step freezing procedures. However, fertilization rates of cryopreserved sperm were significantly lower than the controls (P<0.05). The results of this study indicate that high fertilization rates of striped catfish eggs can be achieved using cryopreserved sperm when frozen at 10°C min?1 in DMSO or DMA with either 0.9% NaCl or C‐F HBSS.  相似文献

ABSTRACT

Striped catfish (Pangasianodon hypophthalmus) fingerlings were stocked (60 fishes/m3) in cemented square tanks for ornamental fishery purposes at the College Fish Farm in Mangalore. A total of 400 fishes with a mean weight (g) ranging from 3.24 ± 1.21 to 6.70 ± 1.13 and a mean length (cm) of 7.50 ± 0.94 to 9.50 ± 1.10 were examined. A severe outbreak of salt-like granule white spots was found on the body surface. Ichthyophthirius multifiliis, a ciliate pathogen, was identified as the causative agent by clinical signs, wet mount, and histopathological observations. Infected fishes were transferred and equally distributed to the 0.45 m3 glass aquaria and treated with three treatments: (T1) methylene blue + salt; (T2) raising temperature with salt; and (T3) formalin + malachite green. The best fingerling survival (55 ± 9.36%) was obtained by elevated water temperature with salt in T2.  相似文献

A bacterial infectivity challenge model of Edwardsiella ictaluri in striped catfish was developed. All experiments were conducted using a bacterial isolate of E. ictaluri that had been recovered during a natural outbreak of bacillary necrosis of Pangasianodon (BNP) in farmed striped catfish Pangasianodon hypophthalmus in Vietnam. Time of immersion in 107 CFU.ml−1 had a significant effect on mortality. The immersion bacterial dose of 107 CFU/ml for 30 s resulted in a cumulative percentage mortality of 63%. Three to four days post-bacterial challenge, fish showed gross clinical signs of natural BNP and E. ictaluri was recovered and identified from these fish. Moreover, a cohabitation challenge was evaluated as an alternative challenge method, although the mortalities among the infected fish were lower at around 15%–40%. This study confirmed the horizontal transmission of E. ictaluri in striped catfish and elucidated that cohabitation challenge could be used in reproducing the disease under controlled conditions.  相似文献

Survival rates of the larvae of sutchi catfish Pangasianodon hypophthalmus are reported to be three times higher under dim conditions (0.1 lx) than those under 100 lx. In this study, larval behaviour of sutchi catfish was examined under various light intensities (<0.01, 0.1, 1, 10 and 100 lx) using a CCD camera to understand why survival rates vary under different light intensities. Five‐day‐old larvae showed significantly higher swimming activity under <0.01, 0.1 and 1 lx than those under 10 and 100 lx. On the other hand, the larvae showed significantly higher aggressive behaviour under 10 and 100 lx; swimming larvae attacked resting individuals more frequently under 10 and 100 lx than those under 0 and 0.1 lx. Aggressive behaviour was considered to induce lesions, inflicted by the sharp teeth of attacking larvae, on larval skin surfaces. It is considered that the chemical substances would generate from injured skin surfaces then acted as stimuli, causing the cannibalistic behaviour in other fish around the injured fish. This study provided evidence that the observed higher survival rates depended on lower frequency of aggressive behaviour under dark or dim conditions. It is therefore recommended that larval rearing of sutchi catfish be conducted under dim (less than 1 lx) conditions.  相似文献

A nutrition trial with striped catfish (Pangasianodon hypophthalmus) juveniles was undertaken to evaluate the effect of replacing dietary fishmeal (FM) protein with corn gluten meal (CGM). A diet with FM as the main protein source was used as the control diet (FM). Five experimental diets (approximately 320 g kg?1 crude protein) were formulated to progressively replace 20% (CGM20), 40% (CGM40), 60% (CGM60), 80% (CGM80) and 100% (CGM100) of FM protein. Fifteen fish per tank (initial weight 11.2 ± 0.6 g) were randomly distributed into 18 80‐litre fibreglass tanks connected to a closed recirculation system (temperature 30.3 ± 1.0 °C). The diets were tested in triplicate for 12 weeks. The final weight and specific growth rate (SGR) of fish fed diets CGM20, CGM40 and CGM60 were not significantly different compared to fish fed the FM diet. Feed intake (FI) tended to decrease with increasing dietary CGM level. Striped catfish fed FM, CGM20 and CGM40 had significantly lower feed conversion ratio (FCR) compared with fish fed CGM80 and CGM100 (< 0.05). The protein efficiency ratio (PER) of fish fed the CGM80 and CGM100 diets was significantly lower than those of all other treatments (< 0.05). Total ammonia‐nitrogen (TAN) excretion increased with elevated dietary CGM inclusion. The viscerosomatic index (VSI) of fish fed the CGM80 and CGM100 diets were significantly higher (< 0.05) than those of fish fed the other treatments. The crude lipid content in the final body composition of the striped catfish was elevated significantly with increasing dietary CGM levels. Fish fed the CGM80 and CGM100 diets displayed haematocrit levels significantly lower (< 0.05) than those fed the other diets. The haemoglobin content in fish was significantly higher in fish fed CGM20 and lower at CGM100 compared to fish fed the FM diet. The results of the present trial indicated that the optimum level of FM protein replacement with CGM determined by quadratic regression analysis was 25.1% on the basis of maximum SGR.  相似文献

Abstract

An on-station pond trial was conducted for 6 months to investigate the suitability of mixed-feeding schedules with varying protein levels on the growth of sutchi catfish, Pangasius hypophthalmuswith silver carp, Hypophthalmicthys molitrixwith a view to minimizing the cost of production. Five different feeding regimes employed were: high protein diet (30% protein, H), low protein (15% protein, L), one day low protein diet followed by one day high protein diet (1L/1H), 7 day low protein diet followed by 7 day high protein diet (7L/7H), and 14 day low protein diet followed by 14 day high protein diet (14L/14H). Ten experimental ponds each of 0.008 ha size were divided into 5 treatment groups each with two replicates. Fishes were stocked at the ratio of 80:20 (sutchi catfish:silver carp) at the total rate of 25,000/ha. Feeding rate was calculated only on the basis of sutchi catfish weight only and fish were fed at the rate of 15,10, 8, and 5% of their body weight for 1st, 2nd, 3rd months, and the rest of the experimental period, respectively. The ranges of water quality parameters during the study period were: temperature 22°C to 32.8°C, dissolved oxygen 4.30 to 7.44 mg/L, pH 6.85 to 7.50, and transparency 21.9 to 32.2 cm. The result of the study showed that significantly (P < 0.05) higher fish weight gain was achieved in feeding schedules of H and 1L/1H and lowest with 14L/14H. The feed conversion ratio (FCR) values ranged between 2.06 and 2.51 with H and 1L/1H showing significantly the best FCR. The FCR values were very low when weight of silver carp was considered which ranged between 1.58 and 1.68. The survival (%) of sutchi catfish were lower (78.1 to 82.5%) compared to that of silver carp (90–95%). The total production of sutchi catfish ranged between 3594 and 7095 kg/ha and silver carp ranged between 1777 and 2220 kg/ha. A simple economic analysis showed that fish in 1L/1H fed alternately with low and high protein diets resulted in the highest net profit. The result of the study suggest that for profitable sutchi catfish farming in Bangladesh, farmers can adopt a mixed-feeding schedule of alternated low and high protein diet instead of continuously feeding a high protein diet.  相似文献

The Mekong giant catfish Pangasianodon gigas is endemic to the Mekong River and is a critically endangered species. The genotypes of the microsatellite DNA (msDNA) and mitochondrial DNA (mtDNA) markers (right domain of the control region) were detected to evaluate the present status of genetic divergence of this species from the Mekong River in Thailand and Cambodia. The observed and expected heterozygosity values of Mekong giant catfish in Thailand and Cambodia were relatively low in comparison with those of other nonendangered freshwater fish species. These two populations from Thailand and Cambodia showed similar levels of genetic diversity, as evaluated by the 384 nucleotides of the mtDNA control region with 13 haplotypes. The pairwise F ST value between the two populations based on the genotype frequencies of msDNA and mtDNA markers suggested a close genetic relationship between the populations in Thailand and Cambodia. The results of this study support the conclusion that the Mekong giant catfish is critically endangered. Care should be taken to sustain the genetic diversity of this species, as the level of genetic variability has already decreased in the wild population. This species is a target species for an ongoing stock enhancement program in the Mekong River in Thailand. It is proposed to apply these markers for proper broodstock management, such as for minimal kinship selective breeding in the hatchery.  相似文献

Farming of the striped catfish, Pangasianodon hypophthalmus, is a major aquaculture activity in Bangladesh, particularly in the district of Mymensingh. However, pangasius farm management practices and the socio‐economic impacts of pangasius farming systems in Mymensingh have not yet been adequately described in the literature. This article provides an overview of the present status and characteristics of pangasius culture in Bangladesh based on data from a study conducted in Mymensingh district during 2009. The mean productivity of pangasius was 36.9 MT ha?1, where 87.9% of the farms produced between 15 and 65 MT ha?1 of pangasius, with an additional 10–20% Indian major and Chinese carps and Nile tilapia. Pangasius aquaculture in Bangladesh has improved the economic and social status of a variety of stakeholders in communities where the fish is farmed.  相似文献
