Twenty-eight Norwegian Red Cattle dairy cows were fed silage ad libitum and restricted amounts of concentrates. Blood samples were collected before morning feeding, once or twice weekly, from 2 weeks before to 12 weeks after calving. Parameters of liver function, carbohydrate status and fertility were recorded in order to assess their interrelationships. Eight cows were treated for clinical ketosis. Four of these had to be treated 2 or 3 times. Aspartate aminotransferase and bilirubin showed the highest within-animal coefficients of correlation with acetoacetate. Analysis of variance revealed a significant effect of carbohydrate status (indicated by plasma acetoacetate levels) on the levels of aspartate aminotransferase, glutamate dehydrogenase and sorbitol dehydrogenase, though only a small part of the total variation was explained by this factor. The estimated volume density of liver fat in the 4th week of lactation averaged 6.0 +/- 6.4% (+/- SD) ranging from 0.1-25.1%. Liver fat content at this stage of lactation was not significantly correlated with other indicators of liver function or carbohydrate status. Cows treated for clinical ketosis had significantly lower plasma progesterone values at the time of first ketosis treatment than untreated multiparous cows. The frequency of high progesterone values (greater than 3 ng/ml) being significantly lower in treated than in untreated cows during the period from 3-5 weeks post partum, though not at later stages. In conclusion, the results revealed a significant relationship between carbohydrate status and liver function, and also between clinical ketosis and luteal function.  相似文献

Serum bile acid (SBA) reference intervals were established by use of a radioimmunoassay method for fasting dogs to be 0.2 to 4.3 micro mol/L (n = 60) and for 2 hour postprandial samples to be 0.6 to 24.2 micro mol/L (n = 37). The SBA reference intervals estimated using an enzymatic method were 0 to 8.6 micro mol/L for fasting (n = 26) and 0 to 29.8 micro mol/L for 2 hour postprandial samples (n = 36). The correlation between the two methods including samples from healthy dogs and clinical cases is good (n = 128, r = 0.82, p < 0.0001). The radioimmunoassay method is linear to 50 micro mol/L and the enzymatic method is linear to 100 micro mol/L, thus both methods require serum dilutions to be made in many cases of primary liver disease. The enzymatic method is less expensive and more convenient for use in a clinical laboratory but requires a greater sample volume (400 micro I) than the RIA method (50 micro I). Both methods have adequate precision and accuracy to be useful as diagnostic tests of liver function in dogs.  相似文献

Fasted Brazilian squirrel monkeys (BrSMs) exhibited slightly higher serum bilirubin levels (0.30±0.05 mg/dl) than others in the fed state (0.13±0.01). The mean liver weight was 50% lower following a 22 h fast. The rate of bile flow was unaffected by fasting and averaged 13.8 l/min/kg and 47.5 l/min/100g liver in six BrSMs. No significant difference in mean bilirubin excretion/min was observed on a body weight basis following fasting. When the mean rate of bilirubin excretion was calculated as a function of liver weight, a two-fold higher rate was present in fasted monkeys, but only at the p=0.06 level of statistical significance. From data collected in this and earlier studies, it would appear that BrSMs represent the best animals studied to date to serve as experimental controls in comparative studies with Bovilian squirrel monkeys which exhibit a Gilbert-like syndrome.  相似文献

An intravenous bile acid loading test was performed on 16 goats using various dosages and sampling protocols. A recently developed enzymatic bile acid assay provided an economical and technologically simple method for bile acid analysis. Intravenous injection of bile salts into goats was well tolerated. Clearance times were rapid. A protocol for performance of the test was suggested and a practical reference range for T 1/2 clearance of 4+/-1 minute was determined. Further investigations of this test in the assessment of liver function in the ruminant appear warranted.  相似文献

We cloned and expressed bile salt hydrolase gene of Lactobacillus plantarum M1-UVS29 in Lactococcus lactis NZ9000 successfully. Gene-specific primers for amplification of L. plantarum bsh were designed by using sequence which availabled from Gen Bank. The production of PCR amplicon was confirmed by sequencing and cloned into pMD18-T vector, and then recombined into expression vector p NZ8148 and yielding vector p NZ8148-BSH. p NZ8148-BSH was transferred into Lactococcus lactis NZ9000. Sequencing indicated that the cloned bsh fragment contained 995 nucleotides, and shared 99.3% sequence homology with bsh gene from L. plantarum MBUL10. Cloned bsh fragment was successfully transduced into NICE expression system and confirmed by PCR and restriction digest. Recombinant BSH protein was analyzed by SDS-PAGE. The molecular weight of BSH protein was approximately 37 ku. Activity of the expressed protein was 0.77 μmol · min-1. The successfully expressed proteins by genetic engineering technology made the function of lactic acid bacteria be abundant and laid the foundation for further researches into cholesterol-lowering lactic acid bacterium food and probiotics.  相似文献

为研究饲料中添加胆汁酸对红鳍东方鲀脂肪酸组成及抗氧化能力的影响,以鱼油为主要脂肪源,配制五种等氮饲料,以初始体重14g的红鳍东方鲀进行为期56天的投喂养殖实验。设置适宜脂肪含量(8%)的对照组,并在对照组添加0.02%及0.10%的胆汁酸,分别获得低胆汁酸添加组(BA)和高胆汁酸组(HBA)。另设一高脂组(脂肪含量12.5%,HL组)及高脂+0.02%胆汁酸组(HLBA组)。结果表明,添加胆汁酸的处理组肌肉和肝脏MUFA均低于对照组,其中HBA组显著低于对照组(P<0.05)。肌肉中n-6PUFA含量在HLBA和HL组显著低于其他组(P<0.05)。在适宜脂肪含量的三个处理组间,肝脏中的C20:0、C16:1n-7、C18:1n-9、C18:2n-6、C20:4n-6、C18:3n-3、C18:4n-3、C20:5n-3和C22:6n-3都呈现随着胆汁酸添加量增加而降低的趋势。血清超氧化物歧化酶活性和谷胱甘肽过氧化物酶活性都有随着胆汁酸添加而升高的趋势,且在高脂组显著高于对照组(P<0.05);血清谷胱甘肽还原酶则呈现相反的趋势。肝脏中谷胱甘肽过氧化物酶1的mRNA相对表达量在对照组显著高于其他组,而BA组氧化还原酶1的基因表达量显著高于对照组;在高脂组添加胆汁酸显著升高了过氧化氢酶的基因表达量。综上所述,饲料中胆汁酸的添加降低了饲料及鱼体组织中一系列长链脂肪酸的含量;胆汁酸对血清抗氧化蛋白活性的影响与其对肝脏抗氧化相关基因表达的影响结果不一致。  相似文献

【目的】对从健康仔猪粪便中筛选分离出一株发酵乳杆菌的益生性能进行测试,为其在畜禽动物养殖中的应用提供参考。【方法】采用Rogosa SL完全选择性培养基分离出106株乳酸菌,通过革兰氏染色、酸和胆盐耐受初步筛选出1株潜在的饲用乳酸菌,经16S rDNA序列鉴定为发酵乳杆菌LF1,并对其益生性能潜力进行评估,包括抑菌活性(针对大肠杆菌K88、金黄色葡萄球菌、猪链球菌、沙门氏菌以及弧菌等12种病原菌)、对模拟胃液中酸性环境的耐受性、对胆盐的耐受性以及对小肠上皮细胞的粘附性。【结果】发酵乳杆菌LF1耐酸和耐胆盐的能力较强,显示出较强的益生菌潜力,但LF1菌株对猪肠道上皮细胞的粘附能力不强。在生物拮抗作用方面,发酵乳杆菌LF1的培养上清可以显著抑制多种常见动物病原菌生长。【结论】发酵乳杆菌LF1具有较好的益生性能,通过进一步的动物实验评估后,可以作为应用于畜禽养殖业的益生菌候选菌株。  相似文献

A 9‐week feeding experiment was conducted to investigate the effects of dietary bile acids (BAs) on juvenile genetically improved farmed tilapia (GIFT) (Oreochromis niloticus) based on the evaluations of growth performance and parameters relevant to lipid metabolism. Each of five vegetable protein‐based diets containing BAs at a level of 0, 0.05, 0.15, 0.45 or 1.35 g/kg diet was fed to three replicates with 40 fish (8.2 g per fish). The results showed that weight gain (WG) increased significantly with the increase in BAs from 0 to 0.15 g/kg diet and then decreased significantly at a higher BA supplementation. Dietary BAs significantly reduced the crude lipid content in the whole body, muscle and liver tissue of GIFT. Fish fed diet with 1.35 g BAs/kg diet developed serious nuclear migration and vacuolization in hepatocytes. Gall bladder appeared to contain white solid and has fragile capsules. Dietary BA supplementation had significant effects on serum biochemical indices and activities of lipid metabolism enzymes in liver and intestine. In conclusion, dietary bile acid supplementation (0.15 g/kg) can facilitate the lipid metabolism and therefore promote the growth of tilapia. However, overdosed dietary BAs induced gallstone development, disrupted lipid metabolism and depressed the growth performances of GIFT.  相似文献

SUMMARY: The aim of this study was to investigate the cause of green liver in red sea bream fed substitute protein diets. Red sea bream Pagrus major was given either of the following diets for 28 weeks: (1) control diet (50% fish meal), (2) low level fish meal diet (15% fish meal), and (3) non-fish meal diet (0% fish meal). The green liver was observed in all groups tested, but the incidence was much higher in the experimental diet groups. The feeding of substitute protein diets reduced plasma triglyceride and cholesterol levels. However, there was no significant difference in plasma hepatic enzyme activities and plasma bile salts concentration among the treatments. Fish fed the substitute protein diets showed low hepatic taurine levels with an appearance of a biliverdin in the liver. Moreover, the proportion of ditaurobilirubin to total biliary bile pigments was significantly lower in fish fed the substitute protein diets. These data indicate that feeding of substitute protein diets did not induce any cholestatic hepatobiliary obstructions and that the low hepatic taurine level was one of the probable factors responsible for the occurrence of green liver in red sea bream fed substitute protein diets.  相似文献

[目的]有研究显示草鱼肠道健康与胆固醇代谢具有较为紧密的关系,本研究探讨草鱼肠道损伤对胆固醇合成代谢通路基因表达的影响.[方法]以池塘养殖条件下的草鱼为研究对象,在对肠道损伤进行外观形态、组织切片和血清指标评估的基础上,测定了草鱼血清中的甘油三酯(TG)、高密度脂蛋白(HDL)、低密度脂蛋白(LDL)、总胆汁酸(TBA)和总胆固醇(TC)含量,采用荧光定量PCR(qRT-PCR)方法,定量检测肠道和肝胰脏组织的胆固醇、胆汁酸生物合成酶和调节控制酶的基因表达水平的变化.[结果]肠道损伤的草鱼血清HDL、LDL、TC和TBA的含量显著下降(P<0.05),肝胰脏和肠道组织的胆固醇、胆汁酸合成酶相关基因表达水平极显著上调(P<0.01),与此同时,负调控胆汁酸合成的法尼醇X受体FXR和调控胆固醇逆转运途经的血浆胆固醇酯转移蛋白CETP基因表达水平极显著下调(P<0.01).[结论]草鱼肠道损伤后,胆固醇、胆汁酸的重吸收可能遭到破坏,刺激了肝胰脏和肠道的胆固醇、胆汁酸生物合成强度,从而维持胆固醇在机体内的代谢平衡.  相似文献
