Background: Despite frequent clinical use, information about the pharmacokinetics and efficacy of pantoprazole in camelids is not available. Objectives: To examine the pharmacokinetics of both IV and SC pantoprazole and to determine whether pantoprazole administration would increase 3rd compartment pH in alpacas. Animals: Six healthy adult alpacas. Methods: Alpacas were fitted with a 3rd compartment cannula for measuring gastric pH. After recovery, alpacas received 1 mg/kg pantoprazole IV, q24h for 3 days or 2 mg/kg SC q24h for 3 days. Alpacas received both IV and SC pantoprazole, with a minimum of 3 weeks between treatments. Third compartment pH was recorded and plasma samples were taken for pharmacokinetic analysis. Results: Pantoprazole induced a slow but sustained increase in 3rd compartment pH when given by both the IV and SC routes. Third compartment pH was significantly increased as compared with baseline values (1.81 ± 0.7; mean ± SD) at 24 (2.47 ± 0.8), 48 (3.53 ± 1.0) and 72 hours (4.03 ± 1.3) after daily IV administration of pantoprazole. Third compartment pH increased from 1.73 ± 0.6 at baseline to 3.05 ± 1.1, 4.02 ± 1.4, and 3.61 ± 1.6 at 24, 48, and 72 hours after SC administration, respectively. Pharmacokinetic analysis demonstrated that pantoprazole had a short elimination half‐life (0.47 + 0.06 h) and a high clearance rate (12.2 ± 2.9 mL/kg/min) after both IV and SC administration. Conclusions and Clinical Relevance: Based on the results of this study, pantoprazole represents a safe and effective drug for increasing 3rd compartment pH in camelids. Either IV or SC administration is likely to be an effective treatment for gastric ulcers. 相似文献
A field experiment was carried out in Kolda (southern Senegal) from July 1986 to July 1988. Its goals were to: (1) describe the patterns of mortality of female Guinean goats by age, season and year; (2) assess preventive measures against respiratory diseases and gastrointestinal parasitism in reducing mortality; and (3) estimate the overall impact of these measures on survival to 1 year of age. Preventive measures for respiratory disease included vaccination against peste des petits ruminants (PPR) and pneumonic pasteurellosis (Pasteurella multocida types A and D). Control of gastrointestinal parasites was by deworming does with morantel (7.5 mg kg−1, three times during the rainy season). The effects of vaccines and deworming were tested in a randomised factorial field experiment with villages being the experimental units. A total of 19 villages, 113 goat herds and 1458 goats were included in the study.
Generalised linear models of survival for five cohorts of goats (defined by five different birth seasons) used a binomial assumption for the response distribution and a complementary log–log link. Explanatory variables included age, season, year, vaccination, deworming and their interactions. A complex a priori model was built on the basis of previous epidemiological knowledge; a purposively selected set of simpler models was compared to this full model by the Akaike information criterion (AIC) and derived statistics. Inference on 1-year survival and treatment effects accounted for model-selection uncertainty. It was carried out with a bootstrap procedure and used information from the whole set of selected models.
Large variations in mortality by year and season were observed but no regular seasonal pattern was apparent. Mortality probabilities of kids in dewormed groups decreased quickly after birth, but remained elevated up to 9 months of age in the non-dewormed groups. Deworming lowered the risk of mortality. Vaccination alone was not protective (except during an observed outbreak of PPR). 相似文献
Enteral nutrition coupled with bacterial colonization has been shown to have major functional and developmental effects during the postnatal period of neonates. In this study, we developed a highly sensitive premature pig model to elucidate the specific developmental impact of initial bacterial colonization on premature neonates by comparing germ-free and conventionally reared pigs. Thirty-eight preterm pigs (93% gestation) were delivered via caesarean section and reared in either germ-free or conventional isolators for 40–48 h. Pigs were fed either infant milk formula or sow's colostrum. Enteral feeding for two days had trophic effects on gastrointestinal weights, particularly for pancreas, stomach and small intestine (SI). The absence of bacteria in formula fed pigs resulted in a mucosa that appeared more robust and had higher weights for both SI and pancreas, compared to conventional formula fed pigs (P < 0.05). Colostrum fed pigs also had markedly increased mucosal SI proportions, lung and spleen weights compared to conventional formula fed pigs. Colostrum fed pigs and germ-free pigs were similar although stomach, pancreas and distal SI weights were highest for germ-free pigs. The results demonstrate that the initial bacterial colonization interacts with diet to modulate the early neonatal organ development, particularly of the GIT. 相似文献
The levels of various ions (Na+, Cl−, Ca2+, Mg2+, K+) in the fluid phase of the gastrointestinal contents and of the plasma were determined in four teleosts with different feeding habits: traira (Hoplias malabaricus), hassar (Hoplosternum littorale), silver catfish (Rhamdia quelen), and grass carp (Ctenopharyngodon idella). The results showed that most ionic levels in the fluid phase of the gastrointestinal tract seem to be affected by feeding habit and that these levels can be different from those found in the plasma. In practical terms, these results suggest that ionic levels of the gastrointestinal contents should be considered when preparing solutions for studies of intestinal absorption in fishes. 相似文献
