将健康的围产期奶牛30头随机分为三组,分别于产前28d开始饲喂NRC标准日粮(营养水平100%组,即Ⅰ组)、NRC标准增加20%日粮(营养水平120%组,即Ⅱ组)和NRC标准减少20%日粮(营养水平80%组,即Ⅲ组),产后各组奶牛均饲喂标准泌乳日粮,至产后56d结束,观察干奶期不同营养水平对围产期健康奶牛血清总胆红素、蛋白及转氨酶的影响。试验结果表明,围产期是机体生理状况处于剧烈变化的时期,奶牛产后的蛋白质代谢活动、转氨酶活性较产前明显增强;但健康奶牛均能通过调整糖代谢、脂代谢和蛋白质代谢,促进能量与营养物质间的相互转化,适应机体的各种生理变化。  相似文献

本试验研究了不同断奶日龄仔猪血清代谢物水平的变化。试验设17日龄断奶、21日龄断奶、28日龄断奶、35日龄断奶4个处理组,每个处理3个重复,每个重复1窝,定时采血。结果表明,17、21日龄断奶组仔猪血糖浓度显著低于35日龄断奶组(P〈0.05),在断奶后2~3周时间内,以17日龄断奶组仔猪血糖浓度波动最大;4个不同断奶日龄组之间仔猪血清尿素氮浓度没有显著差异;与35日龄断奶组比,17日龄断奶仔猪血  相似文献

大豆种子7S、11S球蛋白及7S球蛋白亚基的研究   总被引:2,自引:0,他引:2  
利用线性梯度SDS-PAGE方法分析了黑龙江省422份大豆资源材料的7S和11S球蛋白含量、7S球蛋白α′、α和β亚基的含量变化,以及百粒重、蛋白含量、油分含量、11S球蛋白、7S 球蛋白、11S/7S比值的相关性。结果表明大豆种子贮藏蛋白品种间亚基带型基本一致,但是品种间相同亚基含量差异很大,变异系数均大于10%,变异类型丰富,本实验发现1份α′亚基缺失材料。大豆百粒重、蛋白含量、油分含量、11S球蛋白、7S 球蛋白以及11S/7S比值之间的相关性结果表明:11S球蛋白含量和7S球蛋白含量间(r=-1**)、蛋白含量和油分含量间(r=-0.776**)显著负相关;11S/7S 比值和百粒重、蛋白含量以及油分含量间无关.  相似文献

Albumin and globulin fractions, obtained after modifications of the Osborne's fractionation scheme, were investigated in the seeds of three cultivars of Phaseolus vulgaris, after removal of the seed coats, to prevent interactions of tannins and phenols with the globulins. Also, by using an appropriate buffer to separate the globulins more efficiently, no cross-contaminations were observed in SDS-PAGE between these two fractions. The combination of these two procedures resulted in globulin to albumin ratios 70percnt higher than those published previously with the same cultivars. Amino acid compositions of total globulin, phaseolin and albumin fractions are presented. Whereas the albumins contained much more cysteine than the globulins, the concentrations of methionine were evenly distributed in these two fractions. Also, the bulk of methionine was found in phaseolin; cysteine was found in non-phaseolin proteins. In addition, the globulins contained unexpectedly high amounts of lysine.  相似文献

Multigenic glutelins and monogenic globulin are major storage proteins accumulating in vacuole-derived protein body (PB-II) of rice (Oryza sativa L.) seeds. Because their interplay in PB-II formation was scarcely known, the effect of globulin-less mutation on glutelin accumulation was investigated. In globulin-less mutants, no phenotypic defect was found in seed and plant growth, while PB-II was deformed and apparent glutelin composition was changed, producing new glutelin α polypeptides X1–X5. 2D-PAGE of different combinations of globulin-less and glutelin subunit mutations suggested that the X1/X2, X3, and X4/X5 were derived from glutelin GluB1/GluB2/GluB4, GluA3, and GluA1/GluA2 subunits, respectively. Western blot with glutelin GluB4 subunit-specific and its variable region discriminable antibodies indicated at least in part the new spots X1/X2 are partially degraded products of GluB4 α polypeptides by the removal of 2–39 residues from C-terminus. Time course experiments with maturing seeds indicated the partial degradation of GluB4 occurred earlier (from 7 days after flowering) and higher than that of GluA1/GluA2. Considering the above results together with the fact that globulin accumulates at the periphery of PB-II and its absence produces deformed PB-II, globulin protects glutelins from proteinase digestion and thereby facilitates stable glutelin accumulation.  相似文献

利用改进的聚丙烯酰胺凝胶电泳(PAGE),对玉米种子清蛋白和球蛋白进行研究.结果用含有NaCl的球蛋白提取液Ⅰ提取5次蛋白质后,清蛋白和球蛋白几乎全部被提取;而用不含NaCl的清蛋白提取液Ⅱ提取4次蛋白质后,再用提取液Ⅰ提取,可得到球蛋白组分,而且在清蛋白中也有相同迁移位置的谱带存在.表明对球蛋白进行提取操作时,总有清蛋白相伴随,而对清蛋白进行提取分析时一般不会有球蛋白的存在;球蛋白的遗传组分较清蛋白少,并在清蛋白中一般都有相同分子量的组分存在。  相似文献

东北大豆品种贮藏蛋白7S和11S组分及其亚基相对含量分析   总被引:3,自引:0,他引:3  
利用聚丙烯酰胺凝胶电泳分析了黑龙江、吉林、辽宁和内蒙古4省(区)近10 a来通过审定的163个大豆品种的11s和7s球蛋白亚基组成、相对百分含量和11s/7S比值,并对其进行了省份问比较分析和相关性分析.结果表明:品种问大豆蛋白各亚基的相埘含量存在较大差异;163份供试材料的7s、11S球蛋白平均相对含量分别为28.95%和56.30%.变幅分别为18.40%~36.20%和47.90%~71.50%,11S/7S比值的变异幅度在1.34~3.32之间,平均值为1.97;初步筛选出α和A3缺失或稀少的特异大豆品种9份;吉林、黑龙江、辽宁和内蒙古4个地区大豆群体蛋白亚基组成及11S/7S比值存在显著差异.7S和11S组分含量问存在极显著的负相关(r=-0.2862,P<0.01).东北大豆品种贮藏蛋白7S和11S组分及其亚基相对含量存在显著变异,为大豆品质育种和豆制品加工业原料选择提供了丰富的物质基础.  相似文献

The effect of two rice endosperm proteins, glutelin and globulin, on the physicochemical properties of rice starch and flour was investigated. Albumin, globulin, prolamin and glutelin were sequentially extracted from defatted rice flour with de-ionised water, 1.5 M NaCl, propan-2-ol and 0.1 M NaOH, respectively, followed by dialysis and lyophilisation. Globulin and glutelin were then added to pure rice starch at various concentrations, separately and together, and the pasting and textural properties of mixtures were analysed by the Rapid Visco Analyser (RVA) and TA-XT2 textural analyser, respectively. The presence of glutelin in rice starch caused an increase in pasting temperature but a decrease in the viscosity parameters of the starch paste. The concentration of glutelin was also positively correlated with the hardness and adhesive properties of the starch gel. The presence of globulin, on the other hand, resulted in a decrease in all the pasting and textural parameters except gel hardness and the changes were linearly correlated with the concentration of the protein for most of the physical parameters. When the two proteins were added to rice starch together, the outcomes in pasting and textural properties were generally dependent upon the relative concentrations of the two proteins, but were also influenced by the presence of the other two protein fractions, albumin and prolamin. The presence of globulin initially accelerated the rate of water absorption by starch during cooking while the presence of glutelin slowed it down, but in both cases, the ultimate amount of water absorbed was significantly lower than that by pure starch. The contrasting effects of the different protein fractions mean that it might be possible to manipulate the textural properties of rice starch and flour to achieve desirable sensory outcomes by varying the proportions of the protein fractions in product formulations.  相似文献

从省内各地区收集53个栽培大豆品种,按Appu Rao和Narasinga Rao的方法,分离鉴定各品种种子蛋白中球蛋白的数量。结果表明,黑龙江省栽培大豆品种种子球蛋白含量占种子蛋白质含量的34.54—80.77%,含量的变幅较大,表明品种间有明显的差异。其中球蛋白含量在60%以上的有11个品种,占品种总数的20.75%,含量在50%以上的品种有22个,占41.51%,还有20个品种其含量低于50%,占品种总数的32.1%。 大豆种子蛋白中的球蛋白占绝大部份。各品种球蛋白含量明显影响豆腐产品的数量。品种的球蛋白与豆腐湿重呈显著正相关,回归分析表明,豆腐湿重对球蛋白存在着线性关系。豆腐干重与球蛋白组分7S/11S值达显著水平,回归分析表明,豆腐干重对7S/11S的回归分析存在着线性关系。  相似文献

The storage proteins of the pili nut (Canarium ovatum, Engl.), quantitatively extracted using the modified Osborne protein fractionationscheme, revealed that the salt-soluble globulin was the main storage proteinwithin the kernel (i.e., >>60.3%). Further physicochemicalcharacterization of this aqueous soluble globulin revealed that it existed in an11S like form and was composed of two main subunits of 22,600 and31,600 Da. These subunits were found to be disulfide-linked (in a 1:1ratio) forming intermediary subunits (i.e., dimers) with a molecular weightof approximately 52,600 Da. The overall molecular weight of the 11Sglobulin was determined to be approximately 300,000 Da suggesting thatthe globulin possessed a dodecameric-like structure of 6 dimers for a totalof 12 subunits. Using differential scanning microcalorimetry, thedenaturation temperature of the globulin was shown to occur at89.3 °C. Overall, the pili nut 11S globulin was found to possessmany similar physicochemical properties to those of other 11S oilseedglobulins.  相似文献
