Abstract: The red cell parasites formerly known as Haemobartomlla and Eperythrozoon spp have been reclassified as hemotrophic mycoplasmas (hemoplasmas) based on strong phylogenetic evidence and 16S ribosomal RNA gene sequences. The latter form the basis for polymerase chain reaction assays used to detect infection. Candidatus designation was given to incompletely characterized species. Like other mycoplasmas, hemoplasmas are small epicellular parasites that lack a cell wall and are susceptible to tetracyclines; their circular, double-stranded DNA encodes only those gene products essential for life. Diseases caused by infection with hemoplasmas range from overt life-threatening hemolytic anemia to subtle chronic anemia, ill-thrift, and infertility. In addition, the organisms may act as cofactors in the progression of retroviral, neoplastic, and immune-mediated diseases. Intimate contact of hemoplasma organisms with RBCs leads to cell injury through immune-mediated and other mechanisms that have not yet been defined. Despite an intense immune response and even with antibiotic treatment, infected animals probably remain chronic carriers after clinical signs have resolved. 相似文献

BACKGROUND: The goals of this study were to develop and apply conventional (c) and real-time TaqMan polymerase chain reaction (PCR) assays for Mycoplasma haemofelis (Mhf), 'Candidatus Mycoplasma haematoparvum' (Mhp), and 'Candidatus Mycoplasma haemominutum' (Mhm) to blood samples of cats to determine the epidemiology of these infections in cats. HYPOTHESIS: Cats are infected with >2 hemoplasma species, and organism load correlates with disease induced by these organisms. ANIMALS: Blood samples from 263 anemic and nonanemic cats were used. METHODS: A retrospective study was conducted. RESULTS: Forty-seven (18%) samples were positive. Three samples (1%) yielded 170 base pair cPCR products, 1 of which was positive for Mhf using real-time PCR. Forty-four samples (17%) yielded 193 base pair cPCR products, 40 of which were positive for Mhm using real-time PCR. Organism loads ranged from 375 X 10(6)/mL to 6.9 x 10(6)/mL of blood. Sequencing of cPCR products from samples testing negative using real-time PCR identified 2 Mhp-like sequences, 1 Mhm-like sequence, and 1 sequence resembling 'Candidatus Mycoplasma turicensis'. Cats infected with Mhm were less likely to be anemic than uninfected cats. Older age, outdoor exposure, feline immunodeficiency virus (FIV) seropositivity, cutaneous squamous cell carcinoma (SCC), and stomatitis were associated with Mhm infection. Cats from the Sacramento Valley were more often infected with Mhm than cats from the San Francisco bay area. CONCLUSIONS AND CLINICAL IMPORTANCE: Cats may be infected with 4 hemoplasma species. The association between Mhm infection, FIV, and SCC may reflect outdoor roaming status of infected cats. The clustered distribution of infection suggests an arthropod vector in transmission. 相似文献

为了快速准确检测华南虎感染的血巴尔通体种类,我们根据已发表的猫血巴尔通体的16S rRNA基因序列,利用软件Primer Premier 5.0设计2对特异性引物,以华南虎源血巴尔通体基因组DNA进行PCR检测。2对引物分别扩增出1452 bp和170 bp的rDNA序列,1452 bp序列与GenBank~(TM)发表的3种猫血巴尔通体序列相似性分别为99.3%、91.7%和79.5%,而170 bp序列与序列AM748929的相似性为100%。实验结果表明华南虎感染的血巴尔通体为大型猫血巴尔通体,实验建立的PCR检测方法为华南虎血巴尔通体病的诊断及分子流行病学的调查提供了新的手段。 相似文献

BACKGROUND: Administration of tetracyclines or fluoroquinolones is associated with improvement in clinical and laboratory abnormalities in cats infected with Mycoplasma haemofelis. No treatment protocol has consistently eliminated the organism, and antimicrobial susceptibility may vary among M. haemofelis isolates. Continued search for effective therapies is warranted. HYPOTHESIS: Marbofloxacin administered at the onset of clinical illness will be safe and effective for the treatment of M. haemofelis. ANIMALS: Fourteen young adult, laboratory-reared cats housed together in a specific pathogen-free facility. METHODS: Twelve cats were inoculated IV with 2.0 mL of blood from 2 M. haemofelis positive cats. Clinical parameters were assessed daily. CBC and hemoplasma polymerase chain reaction (PCR) assay were performed before inoculation, weekly for 1-3 weeks postinoculation (PI) and twice weekly 3-6 weeks PI. Treatment with marbofloxacin (2.75 mg/kg PO daily for 14 days) was initiated in 6 randomly selected cats when PCV was <30% or fever was >102.5 degrees F (39.2 degrees C). Cats that were PCR positive on day 7 of therapy were treated for 28 days. Cats that were PCR negative on day 42 PI were treated with 20 mg/kg methylprednisolone acetate IM on day 50 PI. RESULTS: Significant differences between groups on some days after inoculation included higher PCV and red blood cell counts, lower mean cell volume, and higher mean cell hemoglobin content in marbofloxacin-treated cats. No differences in PCR assay results were noted between groups. CONCLUSIONS AND CLINICAL IMPORTANCE: Marbofloxacin was safe and resulted in more rapid hematologic improvement in M. haemofelis-infected cats, but did not change clinical scores and did not consistently eliminate infection. 相似文献
