[目的]建立北柴胡药材的HPLC指纹图谱研究方法。[方法]采用Zorbax SB-C18(250 mm×4.6 mm ID,5μm)色谱柱,在柱温25℃、检测波长220 nm、流速为1.0 ml/min的条件下,以乙腈-0.05%磷酸水溶液为流动相对9个不同产地的柴胡样品进行梯度洗脱,分析其指纹图谱。采用国家药典委员会出版的《中药色谱指纹图谱相似度评价》(2004年A版)软件,对9个不同批次的北柴胡药材指纹图谱进行相似度分析。[结果]各批北柴胡药材中均有11个特征峰,各峰分离度良好,各批次药材间共有峰的相对保留时间RSD均<1.0%,药材间相似度均>90%。[结论]该研究采用HPLC法,以柴胡皂苷a为内参比峰,建立了北柴胡药材的HPLC指纹图谱,方法稳定、重现性好,可有效控制北柴胡药材的质量。  相似文献

目的:研究乳康胶囊提取工艺。方法:以干膏收率、柴胡皂苷a与柴胡皂苷d总量为指标,采用L(934)正交试验优选最佳提取工艺条件。结果:影响提取工艺因素依次为:B第1次加水量>D第2次加水量>A第1次提取时间>C第2次提取时间。最佳工艺条件为:第1次1.5 h,加水8倍量,第2次提取1 h,加水6倍量。结论:该工艺简便、可行,适合于工业生产。  相似文献

Abstract

Two Bupleurum falcatum cultivars, which originated from Jeongseon, Korea and Kumamoto, Japan were grown for two years to clarify the dry matter (DM) production, efficiency for solar energy utilization (Eu) and saikosaponin a,c and d contents in Fukuoka, Japan. Aerial parts, root and total DM production differed with the cultivar both in the first and second years. Generally, DM production correlated with leaf area index, and there was a significant relationship between total DM and Eu in the plants of the first year in both cultivars. High Eu was observed at the flowering stage in both cultivars in both years. The maxium Eu was 1.02-1.23% in the first year and 1.16-2.09% in the second year. Total saikosaponin content was higher in the first year than in the second year. Although the total saikosaponin content was higher in Jeongseon cultivar than in Kumamoto cultivar in the second year, no difference betweeen the two cultivars was observed in the first year. We concluded that annual cultivation with a high saikosaponin content was preferable for the high yield of saikosaponin.  相似文献

针对北柴胡新类型主要农艺性状及品质性状进行比较,调查各类型株高、茎径、叶长、叶宽、叶长宽比、根径、根长、根质量等主要农艺性状;分析单根干质量、柴胡皂苷质量分数、单根柴胡皂苷收量的动态变化。结果表明:各类型间主要性状差异显著。CY(长叶型)株型较高,叶最长;KY(宽叶型)叶宽明显;CY和KY单根干质量、柴胡皂苷收量高;ZY(中间型)茎径和根径最小,柴胡皂苷质量分数高。北柴胡表型变异引起药材化学成分变化,可通过筛选优良变异类型,有针对性地培育优良北柴胡栽培品种。  相似文献

A competitive enzyme-linked immunosorbent assay (ELISA) for saikosaponins was established using monoclonal antibody (MAb) 3G10. Hybridoma 3G10 prepared by fusing splenocytes immunized with saikosaponin a-BSA (SSa-BSA) conjugate and a hypoxanthine-aminopterin-thymidine (HAT)-sensitive mouse myeloma cell line, P3-X63-Ag8-U1, secreted monoclonal antibodies with wide cross-reactivity to saikosaponins including saikosaponin b(2) (SSb(2)), c (SSc) and d (SSd), which are stereo and/or regio isomers of SSa. The method, at an effective measuring range of 0.6 mug /ml to 2.3 mug/ml of SSa, successfully detected total saikosaponins in Bupleuri radix and Kampo medicines prescribed with Bupleuri radix. Good correlation between ELISA and HPLC analyses of total saikosaponin in a crude extract of Bupleuri radix was obtained after hydrolysis of acyl saikosaponins by treatment with a mild alkaline solution.  相似文献

AIM: To investigate the inhibitory effect of saikosaponin a (SSa) on pentylenetetrazole (PTZ)-induced activation of hippocampal astrocytes in mice. METHODS: Hippocampal astrocytes were isolated and cultured. The cells were randomly divided into control group, PTZ group, PTZ+0.625 mg/L SSa group and PTZ+1.25 mg/L SSa group. The cells were identified by detection of glial fibrillary acidic protein (GFAP). The cell viability was measured by MTT assay. The cell cycle was analyzed by flow cytometry. The expression of GFAP and connexin 43 (Cx43) was mea-sured by ELISA. The level of apoptosis was determined by flow cytometry and Hoechst 33258 staining. RESULTS: The primary hippocampal astrocytes grew by adherent culture, and the processes of the astrocytes were obvious. Immunofluorescence showed positive GFAP expression in the astrocytes. Compared with control group, the viability of the cells and the percentage of the cells in G2/M phase in PTZ group were significantly increased (P<0.05), and the expression of GFAP and Cx43 was also markedly increased (P<0.05). Compared with PTZ group, the viability of the cells and the percentage of the cells in G2/M phase were obviously decreased in PTZ+0.625 mg/L SSa group and PTZ+1.25 mg/L SSa group, and the expression of GFAP and Cx43 was also reduced, whereas the percentage of apoptotic cells was significantly increased (P<0.05).CONCLUSION: SSa significantly suppresses PTZ-induced activation of hippocampal astrocytes, inhibits the cell proliferation and induced apoptosis.  相似文献

窄竹叶柴胡(Bupleurum marginatum var.stenophyllum)为柴胡属多年生高大型草本植物,以根入药称为藏柴胡,具有较高的药用价值。本试验采用实时荧光定量PCR技术,利用geNorm、NormFinder及BestKeeper3种不同数据分析软件,从5个常用的候选基因(Actin,α-tubulin,β-tubulin,Cyclophilin,EF-1α)筛选出可在窄竹叶柴胡不同器官稳定表达的基因作为内参基因,并利用筛选得到的内参基因分析皂苷合成关键酶基因(HMGR,IPPI,FPS,SS,β-AS)在窄竹叶柴胡不同器官的相对表达量。结果表明β-tubulin基因可作为窄竹叶柴胡不同器官稳定表达的内参基因;皂苷合成关键酶基因在窄竹叶柴胡的不同器官中表达量有显著差异,IPPI基因在种子中表达量最高,其他基因在根中的表达量均高于在其他器官中的表达量,其中HMGR,FPS,SS在侧根的表达量高于主根。因此,窄竹叶柴胡各器官皂苷合成关键酶基因的表达量不同,最适内参基因为β-tubulin基因,本研究为窄竹叶柴胡分子生物学研究提供一定依据,促进其合理开发和利用。  相似文献
