Rhinoceros populations in Africa are under severe threat as a result of surging poaching rates and risk-mitigation strategies are continuously adapted in an attempt to ensure the survival of the species. This study compared faecal glucocorticoid metabolite (fGCM) levels of two age classes of limited free-ranging female white rhinos with fGCM levels of adult free-ranging female white rhinos. Subsequently, fGCM alterations in the limited free-ranging animals were monitored following routine dehorning as a measure of the animals’ short-term physiological stress response. Baseline fGCM levels differed significantly between tested groups, with both free-ranging and limited free-ranging adult animals showing significantly higher fGCM levels compared with limited free-ranging juvenile females. In contrast, baseline fGCM levels did not differ significantly between limited free-ranging and free-ranging adult individuals. Routine dehorning procedures resulted in a short-term stress response expressed by a significant increase in fGCM levels 48 h post-dehorning, with stress steroid levels returning to pre-dehorning concentrations 72 h after the procedure.  相似文献

AIM:To further elaborate the effect of glucocorticoid receptor(GR) decrease on level of inflammatory media in rats after critical scald.METHOD:The changes of phospholipase A2(PLA2) activity and concentrations of tumor necrosis factoralpha (TNFα) and malondialdehyde (MDA), the product of lipid peroxidation metabolism in plasma and tissue homogenate have been studied in scalded rats with or without GR blockade by RU38486.RESULTS:The PLA2 activity and the concentrations of TNFα and MDA in plasma and homogenate of pulmonary and renal tissue in scalded rats were significantly higher than those in the controls(P<0.01) and further increased in scalded rats accompanied with GR blockade (P<0.01 or P<0.05).CONCLUSION: Blockade of GR might aggravate the increase of inflammatory mediator caused by scalding. It suggested that GR might potentiate anti-inflammation in scald.  相似文献

AIM: To investigate the functions of GR in the course of hepatic secondary injury after severe multiple injury. METHODS: Rat model was produced by adopting severe thoracic impact injury accompanied with mono-side femur fracture, and glucocorticoid receptor was blocked before severe multiple injury. Hepatic macropathology and alterations under light microscope were examined. Maximal binding volume of glucocorticoid receptor (GR) in hepatic tissue was assayed by radio-ligand binding assay and protein content was assayed by Western blot. RESULTS: Maximal binding volume and protein content of GR were gradually decreased in hepatic tissue after severe multiple injury, obviously lower than that in normal control at 4 h after trauma (P<0. 01), and the lowest value was at 12 h after trauma. Maximal binding volume was 12. 9% of normal control (P<0. 01), and protein content was 21. 9% of normal control (P<0. 01). In addition, no significant pathological alteration in liver was found after trauma. However, the pathological analysis showed that the administration of GR blocker before severe multiple injury could cause severe hepatic congestion and infiltration of inflammatory cells in hepatic sinusoid in a dose-dependent manner. CONCLUSION: GR insufficiency could cause secondary hepatic injury in early stage after severe multiple injury, implying an important role of GR in injury and anti-injury mechanism of hepatic tissue cells.  相似文献

研究建立了一种同时检测饲料中泼尼松、甲基泼尼松龙、倍氯米松、氟氢可的松、醋酸可的松等5种糖皮质激素的液相色谱分析方法,采用乙腈作为提取溶剂,减少了杂质溶出;同时优化色谱条件,减少了背景干扰。选择Agilent Poroshell 120 SB-C18色谱柱(3.0 mm×150 mm,2.7μm),以40%乙腈水为流动相、流速0.60 mL/min、柱温35.0℃、紫外检测器(TUV)、波长254 nm进行检测,结果表明,5种糖皮质激素分离良好,6 min内即出峰完毕,线性关系良好,相关系数为0.99957~0.99996,回收率为83.80%-94.04%,检测限为0.03~0.05 mg/kg。整个试验过程只需提取、离心、吹干、定容,免除了净化步骤,达到了高效、灵敏、低消耗的效果,能满足样品分析要求。  相似文献

旨在研究牛磺鹅去氧胆酸(taurochenodeoxycholic acid,TCDCA)对糖皮质激素受体(glucocorticoid receptor,GR)的活化作用,进而阐明TCDCA作用机制。采用分子对接模拟TCDCA与GR的结合情况,并通过构建含有GFP标签的GR真核表达质粒,经瞬时转染HEK-293T细胞,观察TCDCA对GR核移位的影响。结果表明TCDCA(1mmol/L)可与GR发生结合并激活GR,产生核移位,证明TCDCA抗炎免疫调节作用与其活化GR信号通路相关。  相似文献
