[目的]对生活中常见的几种十字花科蔬菜中芥子碱硫氰酸盐含量进行测定.[方法]建立高效液相色谱检测芥子碱硫氰酸盐的条件,并采用高效液相色谱法对十字花科蔬菜白萝卜、红萝卜、青萝卜、紫甘蓝、西兰花、花椰菜、大白菜、小白菜、卷心菜、油菜、油麦菜中的芥子碱硫氰酸盐进行定性、定量分析,并初步考察白萝卜中芥子碱硫氰酸盐稳定性.[结果] HPLC结果显示,几种十字花科蔬菜中白萝卜、红萝卜、青萝卜、紫甘蓝、西兰花、花椰菜、油菜、油麦菜中均含有芥子碱硫氰酸盐,其中白萝卜中芥子碱硫氰酸盐含量最高(0.006 7%),紫甘蓝中芥子碱硫氰酸盐含量次之(0.002 6%),红萝卜、青萝卜、西兰花、油菜、油麦菜、花椰菜中虽然含有芥子碱硫氰酸盐但含量较低超出检测范围,大白菜、小白菜、卷心菜中未检测到芥子碱硫氰酸盐.[结论]芥子碱在十字花科蔬菜中含量差别较大,白萝卜中的芥子碱硫氰酸盐含量最高.  相似文献

AIM:To investigate the effects of sinapine, an effective monomer of Chinese medicine, on hydrogen peroxide (H2O2)-induced adipogenic differentiation of bone marrow mesenchymal stem cells (BMSCs).METHODS:The undifferentiated rat BMSCs were identified and screened by flow cytometry. The adipogenic differentiation of BMSCs was induced by H2O2, and the toxicity of sinapine on BMSCs was tested by CCK-8 assay. After the modeling method and the concentration range of sinapine were determined, the lipid droplets in the cells were detected by Oil Red O semi-quantitative assay, and the optimal drug concentration was selected. Finally, Oil Red O assay was observed 24 h after drug intervention, and the expression of adipogenic differentiation-related proteins, adipocyte protein 2 (aP2), peroxisome proliferator-activated receptor γ (PPARγ) and glucose transporter 4 (Glut4), at mRNA and protein levels in the BMSCs was determined by qPCR and Western blot.RESULTS:Treatment with H2O2 at 200 μmol/L for 1 h induced BMSCs to differentiate into adipocytes. Below the concentration of 40 μmol/L, sinapine had no toxicity to BMSCs. The best inhibitory concentration of sinapine on adipogenic differentiation was at 15 μmol/L. The number of lipid droplets in sinapine (15 μmol/L) group was significantly lower than that in model group. In sinapine group, the expression of aP2, PPARγ and Glut4 at mRNA and protein levels was lower than that in model group (P<0.01).CONCLUSION:Sinapine inhibits H2O2-induced adipogenic differentiation of rat BMSCs. The mechanism may be related to the PPARγ/AMPK signaling pathway.  相似文献

Sinapine derived from cruciferous plants could be converted into trimethylamine by intestinal microbiota. Its metabolite, trimethylamine N-oxide (TMAO), is closely linked to increased risk of cardiovascular disease and fat deposition in mammals. Hens fed with rapeseed meal (RSM) suffered from fatty liver hemorrhage syndrome (FLHS). This study was conducted to investigate whether RSM-induced fatty liver is due to TMAO via altering microbiota composition and diversity. At 33 weeks of age, 600 laying hens were randomly divided into 5 treatment groups, namely control and 14% RSM treatment groups (DY5, with 16.2% erucic acid [EA] and 74.66% glucosinolate [Gl] contents; MB1, with 3.50% EA and 43.23% Gl contents; DY6, with 6.7% EA and 22.67% Gl contents; XH3, with 44.60% EA and 132.83% Gl contents) for 8 weeks. Results revealed that 3 hens died due to liver hemorrhage after ingesting 14% RSM diet. The 14% RSM decreased serum low-density lipoprotein cholesterol (LDL-C) content (P < 0.01) while tended to increase serum TMAO content compared to the control group (P = 0.08). The 14% RSM diet increased red oil O optical density (P < 0.01), and increased total cholesterol (TC) and LDL-C content in the liver (P < 0.01, and P < 0.01, respectively). The 14% RSM decreased liver total bile acid (TBA) content compared to the control (P < 0.01). The DY6 had a higher TBA content in the liver than the XH3 (P < 0.01). The 14% RSM decreased mRNA abundance of liver X receptors alpha (LXR-α, P = 0.01), and increased mRNA abundance of sterol response element binding protein 2 (SREBP-2, P = 0.04). Results revealed that the in-feed RSM could alter richness and diversity of cecal microbiota compared to the control (P < 0.05). Liver TC content and serum TMAO showed a negative relationship with Proteobacteria and Actinobacteria (P = 0.04). In conclusion, 14% RSM increased liver TC and induced high liver score of FLHS, which was possibly associated with the altered cecal microbiota composition, increased serum TMAO levels and LXR-α and SREBP-2 expressions.  相似文献

[目的]探讨花椰菜芥子碱的提取工艺。[方法]采用单因素试验和L8(27)正交试验设计,考察了提取时间、物料比、提取次数、乙醇浓度、花椰菜粉是否脱脂(脱脂采用索氏提取器,以石油醚为溶剂)、提取温度、提取后是否浓缩共7个因素对提取率的影响。[结果]得出花椰菜芥子碱的最佳热醇提取工艺条件为:花椰菜粉不脱脂,提取时间4.5 h,物料比1∶35,提取次数3次,提取温度70℃,乙醇浓度80%,提取后不浓缩。[结论]结果表明,花椰菜粉在提取前是否脱脂对芥子碱的得率影响最大。  相似文献
