To investigate the pathogens of goat's respiratory disease from a goat farm in Guangxi province, the epidemiological investigation, clinic observation, pathological examination, bacterium isolation and identification, biochemical test, PCR and animal experiment test were conducted.The preventive and control measures were taken on the basis of pathogen epidemiological characteristics and drug sensitive test results.The results showed that Mycoplasma, influenza virus and parasite were negative by isolation and culture or PCR, however, two strains of gram-negative bacteria named MS1 and MS2 were isolated from lung.MS1 was identified as Serratia marcescens by biochemical test and 16S rRNA sequencing, which shared 99% nucleotide homology with other Serratia marcescens in GenBank.And MS2 was identified as E.coil by the same methods that shared 99% nucleotide homology with other E.coli in GenBank.Animal experiment showed that two strains could cause death in mice.The drug sensitive tests showed two strains were highly sensitive to spectinomycin, amikacin, kanamicin and neomycin.Kanamycin and dexamethasone were used to treat the sick goats, and achieved good results.  相似文献

Blood transfusions in veterinary medicine have become increasingly more common and are now an integral part of lifesaving and advanced treatment in small and large animals. Important risks associated with transfusion of blood products include the transmission of various infectious diseases. Several guidelines suggest what infectious agents to screen for in canine and feline transfusion medicine. However, while the risk of bacterial contamination of blood products during storage and administration has not been documented in veterinary medicine, it has emerged as a cause of morbidity and mortality in human transfusion medicine. Clinical experience shows that the majority of blood component bacterial contaminations are caused by only a few species. Unlike other types of bacteria, psychrotolerant species like Pseudomonas spp. and Serratia spp. can proliferate during the storage of blood units at 4°C from a very low titer at the time of blood collection to a clinically significant level (> 105 CFU/mL) causing clinical sepsis resulting from red blood cell concentrate transfusions in human medicine. The purpose of this report was to describe the detection and quantification procedures applied in 4 cases of bacterial contamination of canine and feline blood units, which suggest the need for further investigations to optimize patients’ safety in veterinary transfusion medicine.  相似文献

为明确柞蚕Antheraea pernyi灰卵(工厂化繁育赤眼蜂过程中广泛存在的一种感病寄主卵)对松毛虫赤眼蜂Trichogramma dendrolimi寄生选择与适应性及其繁育子代蜂寄生能力的影响,以柞蚕灰卵作为供试寄主,健康卵作为对照,在无选择和双向选择条件下研究松毛虫赤眼蜂对其的寄生选择与适应性,并比较灰卵和健康卵繁育的子代蜂对0、1、2、3日龄米蛾Corcyra cephalonica卵的寄生能力。结果表明,在无选择条件下,松毛虫赤眼蜂在灰卵上的寄生率、羽化率、单卵出蜂数和总蜂数分别为30.0%、54.1%、39.3头和48.7头,而在健康卵上分别达到96.0%、93.0%、82.5头和96.8头。在双向选择条件下,松毛虫赤眼蜂在灰卵上的寄生率、羽化率、单卵出蜂数和总蜂数分别为33.3%、27.1%、24.7头和52.2头,而在健康卵上分别为68.0%、86.3%、60.6头和74.2头。在无选择和双向选择条件下,松毛虫赤眼蜂在灰卵和健康卵上的发育历期和后代雌性比均不存在显著差异,但灰卵繁育的子代蜂对1、2日龄米蛾卵的寄生数则显著低于健康卵繁育的子代蜂,且后者对米蛾卵的寄生数随着米蛾卵日龄的增加呈逐渐下降趋势,而灰卵繁育的子代蜂对0、3日龄米蛾卵的寄生数要显著高于1、2日龄的。表明柞蚕灰卵会对松毛虫赤眼蜂的寄主选择与适应性以及子代蜂的寄生能力产生不良影响。  相似文献

To investigate the pathogen of bovine respiratory disease complex (BRDC) of a dairy farm in Guangxi, a strain of Mycoplasma and a strain of gram-negative pathogenic bacterium were isolated and identified by the means of field surveys, clinic observation, pathological examination, isolation studies and so on.Treatments were taken according to drug sensitivity test results.The Mycoplasma strain, growing on PPLO medium, formed typical "fried egg" colonies.A 448 bp of oppF fragment was amplified by PCR from the strain and had 98.4% nucleotide identity with Mycoplasma bovis reference isolate PG5 of USA.The biochemical features of the gram-negative bacterial isolate were same with Serratia marcescens.The PCR amplified 16S rDNA of the gram-negative pathogenic bacterium strain was 1 400 bp.It shared 99.0% nucleotide identity with other Serratia marcescens reference strains obtained from GenBank.Animal experiment showed that the gram-negative pathogenic bacterium isolate could cause the mice to die.The drug sensitivity tests showed all isolates were sensitive to spectinomycin, azithromycin, amikacin, gentamicin and neomycin.It was effective to treat with dexamethasone and spectinomycin.Pathogen analysis and drug treatment showed that the BRDC was caused by Mycoplasma bovis and Serratia marcescens.  相似文献

为了将粘质沙雷氏菌(Serratia marcescens)应用在松墨天牛(Monochamus alternatus)防治上,对粘质沙雷氏菌的培养基配方进行了优化。通过单因素试验筛选粘质沙雷氏菌发酵培养基的碳源、氮源和无机盐,利用Plackett-Burman试验设计确定3个主要影响因素,通过最陡爬坡试验,结合Box-Behnken Design试验设计及响应面分析法优化培养基组成成分,获得培养基最佳配方与发酵条件:葡萄糖23.27 g·L-1、蚕蛹粉22.37 g·L-1、酵母粉10 g·L-1、硫酸镁2 g·L-1、磷酸氢二钠3 g·L-1,氯化钠2 g·L-1;在28 ℃、150 r·min-1条件下发酵36 h,装液量体积分数为50 mL/250 mL,接种量4.73 %,pH 7.1~7.3。最后,根据最优培养条件验证发现优化后培养基中生长的菌落数为6.0×109 cfu·mL-1,是初始培养基的9.42倍,与模型预测值相符。研究结果为粘质沙雷氏菌的发酵生产提供了理论基础,为松材线虫病(pine wilt disease)的防治提供了新的途径。  相似文献

粘质沙雷氏菌几丁质酶(ChiC)基因克隆及其生物信息学分析   总被引:6,自引:0,他引:6  
从粘质沙雷氏菌(Serratia marcescensATCC14041)中克隆出几丁质酶基因(ChiC),将回收纯化的PCR产物与载体pMD18-T连接,构建成的重组质粒命名为pMD-Ch iC,将重组质粒转化到受体E.coliDH5α中进行克隆,经BamHI和NheI双酶切验证、核酸序列测定证实,重组质粒pMD-Ch iC含有几丁质酶C基因(ChiC)。利用生物信息学的方法,推测该粘质沙雷氏菌ChiC基因编码的蛋白质由480个氨基酸组成。预测该蛋白的等电点为5.63,分子量约为52kD。针对粘质沙雷氏菌中的几个几丁质酶基因做了进化树,进而验证了粘质沙雷氏菌(Serratia marcescens)几丁质酶(ChiC)在沙雷氏菌几丁质酶中的分类;同时对粘质沙雷氏菌几丁质酶C(ChiC)蛋白的高级结构作出了预测,得到其编码的属于18家族的蛋白质高级结构图谱。  相似文献

[目的]通过对块根芍药内生菌XJU-PA-6产灵菌红素发酵条件的优化,提高其产灵菌红素的能力。[方法]通过单因素试验初步研究碳源、氮源、无机离子、氨基酸、植物添加油及接种量对灵菌红素合成的影响,确定提高灵菌红素产率的最优培养基配方和培养条件。[结果]通过优化试验,菌株XJU-PA-6产灵菌红素最佳发酵条件:40 g/L麦芽糖,20 g/L蛋白胨,0.01 mol/L Mg2+,0.001 mol/L Fe3+,1%脯氨酸,6%葵花油,28℃,180 r/min,振荡培养2 d。在此优化条件下,灵菌红素产量达到290.92 mg/L,比原来提高3.896倍。[结论]优化后的MEA培养基是一种适合内生菌XJU-PA-6生产灵菌红素的优良培养基。  相似文献

[目的]筛选一株高絮凝活性菌株并探究其絮凝特性。[方法]从活性污泥中筛选出一株絮凝剂产生菌,并进行鉴定。对该菌株所产絮凝剂进行提取纯化,并研究其理化性质和絮凝特性。[结果]该菌株被鉴定为沙雷氏菌属(Serratiaplumuthica)。该菌在一定的培养条件下达到最高絮凝活性仅需10h,这有可能大大降低其生产成本。红外分析结果,该菌所产絮凝剂是一种酸性多糖。当微生物絮凝剂用量为0.7mg/L,pH为2-7,温度30-80℃时,均具有很高的絮凝活性。[结论]该菌株所产絮凝剂和其他研究的菌株相比,具有用量少和比较宽泛的pH、温度使用范围的特点。该菌株所产絮凝剂能大大提高活性污泥的沉降能力。  相似文献

粘质沙雷氏菌H30中控制乙偶姻合成的调控因子budR被克隆和鉴定。序列分析表明调控因子budR与液化沙雷氏菌MG1中乙偶姻合成调控因子slaR同源性达80%,属lysR型调控因子。调控因子budR突变可导致粘质沙雷氏菌无法合成乙偶姻,引起培养液pH快速下降和菌体生长缓慢,而在培养过程中维持pH7.0,突变菌的生长可获得恢复,暗示budR的功能为调控粘质沙雷氏菌发酵过程中性产物乙偶姻的合成,以阻止发酵液过度酸化。  相似文献
