洞庭湖水系鳜遗传结构

摘    要:测定了采自洞庭湖水系6个地理位置(湘江常宁、沅江洪江、沅江桃源、澧水石门、洞庭湖万子湖和南岳坡)共100尾鳜(Siniperca chuatsi)的线粒体Cyt b基因DNA序列。结果显示:1 140 bp的Cyt b基因共检测到55个变异位点,100个样本得到9个单倍型。单倍型多样性指数(Hd)和核苷酸多样性指数(Pi)分别为0.777±0.033和0.019 70±0.000 74。洞庭湖的万子湖和南岳坡2个群体遗传多样性指数最高,湘江常宁群体遗传多样性指数最低。分子变异分析(AMOVA)结果显示:FST=0.098 03(P>0.05),9.8%的变异来自群体间,91.2%的变异来自群体内。除常宁群体分化显著外,其它群体没有出现明显遗传分化(两两之间FST在-0.065~0.024之间)。

Abstract:Siniperca chuatsi,an endemic species to China,and its germplasm protection and resources utilization becomes more and more concerned.To know its genetic diversity and differentiation,the mitochondrial DNA Cyt b gene was amplified and sequenced for 100 individuals from six regions of Dongting Lake basin,including Changning,Hongjiang,Taoyuan,Shimen,Wanzihu and Nanyuepo.Altogether 55 variable nucleotide sites existed among the aligned sequences of 1140bp,and 9 haplotypes were found within 100 individuals.The average nucleotide diversity(Pi) was 0.019 7±0.000 74,while haplotype diversity(Hd) was 0.777±0.033.Genetic diversity was the highest in Wanzihu and Nanyuepo population,while that was the lowest in Changning population.AMOVA analysis demonstrated that FST was 0.098 03(P>0.05),small variances occurred among populations(9.8%) and most variances occurred within populations(91.2%).Except Changning population which had significant genetic differentiation,the other populations had no obvious differentiation(Fst among them were between-0.065 to 0.024).
