摘    要:多年生黑麦草成熟胚为外植体,选择CC+2,4-D 4 mg/L为诱导培养基,NB+6-BA 2 mg/L为分化培养基,1/2 NB+IAA 0.5 mg/L+NAA 0.5 mg/L为生根培养基,建立了多年生黑麦草胚性愈伤组织高频诱导与再生系统,确定了G418(150~200 mg/L)、Hpt(50 mg/L)、Basta(20~30 mg/L)的筛选临界浓度,确定了最佳分化时间,探讨了建立多年生黑麦草转化受体系统的必要条件。

Abstract:An efficient regeneration and transformation system for Lolium perenne by using mature seeds of three species was established.An efficient induction and regeneration system from embryonic calli of these species was setup and the conentrations of different antibiotics for callus growth was determined.The essensial requirement of establishing the transformation receptor system for Lolium perenne was discussed in terms of choosing plant tissues,confirming the effect medias and selecting the best regeneration time.
