摘    要:以巴东木莲叶片为材料,通过对反应体系中Mg2+、dNTP、Taq酶、模板、引物浓度等影响因素的优化,确定了一套适于巴东木莲RAPD分析的稳定反应体系,即25μl的总反应体系中:模板DNA 100 ng,Mg2+2.1 mM,引物0.7μM,dNTP0.20 mM,TaqDNA聚合酶3 U。

Abstract:RAPD technology was first employed to study the rare and endangered plant Manglietia patungensis Hu in this paper.An efficient and stable RAPD reaction system for the plant was established through the optimization of the reaction factors:the concentration of Mg~(2+),dNTP,Taq DNA polymerase,template and the primers.The results showed that the optimized RAPD reaction conditions were 100 ng template DNA,2.1 mM Mg~(2+),0.7 μ M primer,0.20 mM dNTP and 3 unit Taq DNA polymerase in a total of 25μl reaction mixture.
