口蹄疫A型病毒AF/72株参考血清的研制

摘    要:目的]研制口蹄疫A型病毒AF/72株参考血清,筛选对实际防疫更具针对性的A型FMD标准血清,为研发安全高效优质的A型FMD疫苗提供实物支撑。方法]用国家口蹄疫参考实验室提供的口蹄疫A型病毒AF/72株的第12代细胞毒AF/72/MF6-BF12,经测定并经Kaber法计算其TCID50为10-8.0/ml;经紫外分光光度法测定其146 S含量,均值为189.0 ng/ml,远大于22.0 ng/ml的国际标准。将此细胞毒与弗氏佐剂联合制成灭活疫苗免疫豚鼠后分离得到血清。结果]经物理性状检验、无菌检验和外源病毒检验,纯净性符合兽用生物制品标准要求;参照国际标准血清的抗体效价测定方法ELISA和微量细胞中和试验检测了此血清的抗体滴度,原血清,强阳性血清和弱阳性血清的中和效价均远小于1∶45,符合标准的要求。结论]对比国际口蹄疫参考血清的抗体滴度确定强阳性血清为口蹄疫A型病毒AF/72株的参考血清,为以后筛选抗原谱广的制苗毒株提供实物支撑。

Preparation of the Reference Serum for the Type A of Foot and Mouth Disease Virus Strains AF/72

Abstract:Objective] The reference serum for type A of foot and mouth disease virus strains AF/72 was researched and the standard FMD serum of type A more targeting actual vaccination was screened for the development of safe,high-efficiency and good-quality type A-FMD vaccines.Method] The TCID50 value of the 12th generation cytotoxicity AF/72/MF6-BF12 of type A of foot and mouth disease virus strain AF/72,which was supplied from the National Reference Laboratory of Foot and Mouth Disease,was 10-8.0/ml with the testing method of Kaber and the contain of 146 S,189.0 ng/ml with the method of UV Spectrophotometric Determination,which was much higher than international standard(22.0 ng/ml).The serum was produced from the immune guinea pig with the inactivated vaccine made from the cytotoxicity united with Freund's adjuvant.Results] The test of physical property,asepsis and exogenous virus indicated that the purity of the product could meet the standard requirement of veterinary biologics.The neutralization ration of efficiency to value of the serum in antibody titers,the original serum,strong and weak positive serum was far less than 1∶45 according to the international standard determination method of serum antibody titer and micro-ELISA test,which was in line with the standard requirements.Conclusion] The strong positive serum was ensured as the reference of the type A of foot and mouth disease virus strains AF/72 international foot and mouth disease through the comparison of the international reference serum with the product in the serum antibody titer,which could provide the support in-kind for the subsequent screening of a broad spectrum of antigen vaccine strain.
