黄曲条跳甲对敌敌畏抗性机理的初步研究

摘    要:采用敌敌畏进行连续2次的黄曲条跳甲种群汰选,以获得中高抗个体,并测定汰选种群和对照种群的药剂敏感性,以及羧酸酯酶、乙酰胆碱酯酶和谷胱甘肽-S-转移酶的比活力及动力学常数.结果表明敌敌畏汰选种群的LC50略高于对照种群,羧酸酯酶的最大反应速度Vmax为0.008 1,米氏常数Km为0.617 9,均明显小于对照种群的最大反应速度(Vmax=0.084 9)和米氏常数(Km=2.369 9);敌敌畏汰选种群的乙酰胆碱酯酶的比活力为0.007 4 OD412· insect-1· min-1,低于对照种群的乙酰胆碱酯酶比活力(0.009 6 OD412· insect-1· min-1),且差异显著;敌敌畏汰选种群的谷胱甘肽-S-转移酶酶比活力为0.005 0 OD340· insect-1· min-1,高于对照种群的谷胱甘肽-S-转移酶酶比活力(0.003 6 OD340· insect-1· min-1),二者差异显著.说明黄曲条跳甲的抗药性与羧酸酯酶、谷胱甘肽-S-转移酶的活性提高以及乙酰胆碱酯酶活性的降低有关.
