摘    要:DNA分子标记技术是检测遗传差异的一项新技术,特别是RAPD标记技术应用更加广泛,DNA质量是保证RAPD分析成功的关键.本文采用改良SDS法对油菜叶片总DNA进行了提取,经琼脂糖凝胶电泳和紫外光光度计分析,结果表明:1、所提取的DNA浓度和纯度都高,可以作为分子生物学研究所用,并为本实验后续的PCR扩增奠定了良好的材料基础;2、不同时期叶片DNA的含量不一样,苗期幼叶DNA含量比青荚期无柄叶DNA含量高.

Abstract:The DNA marker technology is a new technology of measuring the hereditary difference,especially RAPD marker is more extensive in technical application,DNA quality is the key to guaranteeing that RAPD is analysed successfully.This paper adopted SDS improred method to draw the laminae total DNA of the rape, the analized result of the agar-agar candy gel electrophoresis and ultraviolet ray photometer showed that:1. The density and purity of the drawn DNA are all high, it can be used for researchment on molecular biology,and it established a good material foundation for the followed PCR increasing experiment .2.The DNA content of laminae is different in different periods,The DNA content of young leaf in seedling stage is higher than that of sessile leaf in the green period.
