摘 要: | 目的]为蛹虫草各种药理机制及有效成分的研究提供试验依据。方法]在其他条件不变的情况下,分别研究不同酶解液(单一酶和不同的组合酶)、不同菌龄(3、5、7、9、11 d)、不同酶解时间(1、2、3、4、5 h)、不同酶解温度(23、28、33、38、43℃)、不同酶解pH值(pH值4.92、5.29、5.91、6.47、6.98)以及不同渗透压稳定剂(浓度0.6 mol/L的MgSO4、KCl、甘露醇和葡萄糖)对蛹虫草原生质体制备结果的影响,确定制备蛹虫草原生质体的适宜条件。结果]用浓度0.6 mol/L KCl配制成含1.0%纤维素酶和0.5%蜗牛酶的复合酶,在28℃、pH值5.91时,对培养9 d的菌丝体酶解3 h,得到的原生质体数量最多。结论]为以后进行蛹虫草原生质体技术的深入研究和应用奠定了基础。

Research on the Isolation Condition of Militaris Protoplast |

Abstract: | Objective] The basis for the research on the pharmacological mechanism and active ingredients of militaris was explored through the experiment.Method] The effect of the different enzyme solutions(single enzyme and combination of enzymes),the bacteria at different ages(3,5,7,9 and 11 days),the different digesting times(1,2,3,4 and 5 hours),the different digesting temperature(23,28,33,38 and 43 ℃),the different pH values(4.92,5.29,5.91,6.47 and 6.98) and the different stabilizer of osmotic pressure(the concentration of 0.6 mol/L of MgSO4,KCl,mannitol and glucose) on the militaris protoplast isolation was studied under the same other conditions so that the optimal condition of militaris protoplast preparation was determined.Results] The optimal condition for the largest number of militaris protoplast preparation was the enzyme solution of 1.0% cellulase and 0.5% snail enzyme in the concentration of 0.6 mol/L KCl,temperature of 28 ℃,pH value of 5.91,the mycelium at 9 day-age and the time of enzyme treatment of 3 hours.Conclusion] The experiment could promote the future and in-depth study and application of militaris protoplast technology. |
