穿山龙愈伤组织培养研究

摘    要:以穿龙薯蓣的叶柄、茎段、叶和茎尖为材料,对其进行了愈伤组织培养。结果表明:穿龙薯蓣不同外植体(叶柄、茎段、叶、茎尖)均能诱导出愈伤组织,其中以茎尖形成愈伤组织最快,皂甙元含量最高;不同培养基、pH值、接种量、温度、激素等因子对愈伤组织的形成、生长及皂甙元含量有很大的影响;改良MS培养基、2,4-D浓度为3.0~5.0mg/L、BA为1.0mg/L的激素配比使愈伤组织的生长量、产物含量最高。

Abstract:The callus culture on Dioscorea nipponica(leafstalk, stem, leaf, stem tip) was conducted. The results show that the calli induced from different parts have capability to produce Diosgenin which is the secondary metabolism. The stem tip is better than other parts in callus growth and Diosgenin content is influenced by some culture conditions, such as media, pH, temperature and hormone. The optimum cultural conditions on callus culture is incubated in medium with modified medium and 2,4-D(3.0~5.0mg/L) and BA(1.0mg/L),which enables the content of Diosgenin highly and the callus growth quickly.
