二球悬铃木的组织培养和体外植株再生研究

摘    要:以秋季休眠的二球悬铃木枝条的单芽茎段为外植体,接种于MS培养基上。结果表明:MS+6-BA0.5mg/L+IBA0.25mg/L(附加0.6%琼脂+3%蔗糖)最有利于诱导出不定芽,丛生芽诱导期培养温度为28±1℃。MS+IBA0.25mg/L+NAA0.1mg/L(各附加0.6%琼脂+2.7%蔗糖)诱导生根效果相对较好。不定根诱导期培养温度为26±1℃;光照度500~1000lx,光照时间6h/d。

Tissue Culture and in Vitro Plantlet Regeneration of Platanus acerifolia

Abstract:By using single-bud stem segment of Platanus acerifolia ,which is dormant in fall,as materials. They were inoculated into the MS medium. The results showed that,the best medium for adventitious buds induction was MS +6-BA0.5 mg/L + IBA0.25 mg/L,supplemented with 0.6% agar and 3% sucrose. Culture temperature is 28±1 ℃ during induction period of adventitious buds. The optimal medium for adventitious roots induction is MS + IBA0.25 mg/L +NAA0.1 mg/L,supplemented with0.6% agar and 2.7% sucrose. Culture temperature is 26±1 ℃ during induction period of adventitious roots,illuminance of 500~1000lx and illumination time of 6 h/d.
