家蚕糖转运蛋白基因BmST3的克隆及序列分析与表达研究

摘    要:糖转运蛋白在家蚕体内糖类化合物的转运与代谢过程中发挥重要作用。在电子克隆的基础上,通过RT-PCR方法克隆了家蚕糖转运蛋白基因BmST3(GenBank登录号:GQ871756)。生物信息学分析结果显示,该基因位于家蚕27号染色体,开放阅读框(ORF)长1434bp,编码477个氨基酸,预测蛋白有典型的Sugar_tr结构域和12个疏水的跨膜结构域,与埃及伊蚊、致倦库蚊、冈比亚按蚊、赤拟谷盗登录号分别为EAT47626、EDS35465、EAA11457、EFA05337的同源蛋白相似性均在60%以上。RT-PCR检测和基因芯片信号值分析结果显示,BmST3基因的表达具有组织特异性,在家蚕5龄第3天幼虫的9种组织中,主要在马氏管中大量表达,推测其可能在马氏管细胞膜内外物质的跨膜转运中发挥作用。

Abstract:Sugar transporters play important role in the transportation and metabolism of carbohydrates in silkworm (Bombyx mori).BmST3 (GenBank accession No.GQ871756) was cloned by RT-PCR based on sequence obtained via in silico cloning.Bioinformatics analyses indicated that this gene is located on the 27th chromosome.Its open reading frame (ORF) is 1434 bp long and encodes 477 amino acids with a typical Sugar_tr domain and 12 hydrophobic transmembrane domains.A comparison of BmST3 with homologous proteins in Aedes aegypti,Culex quinquefasciatus, Anopheles gambiae,and Tribolium castaneum ( GenBank accession No.EAT47626,EDS35465,EAA11457 and EFA05337 respectively) revealed that their amino acids share sequence similarities of over 60%.It was shown by RT-PCR detections and microarray analyses that BmST3 had a tissue-specific expression pattern.Among 9 tissues detected on day 3 of the 5th instar larvae,the highest expression was observed in Malpighian tubule.It was suggested that BmST3 plays important role in the transmembrane transportation of substances across cell membrane in Malpighian tubule.
