结缕草ZjSGR基因在离体模拟逆境胁迫环境下的功能

摘    要:结缕草(Zoysia japonica Steud.L)是禾本科多年生草本植物,适用于运动场草坪建植等。但结缕草在北方生长绿期较短,制约了其大面积的应用,因此培育绿期长、抗性好的结缕草品种成为重要育种目标。而SGRSTAY-GREEN)滞绿基因的发现为改善结缕草持绿性提供了便利。本研究通过对野生植株与结缕草Zjsgr突变株系的叶绿素含量、MDA (Malondialdehyde,MDA)含量、可溶性蛋白含量和抗氧化酶活性等指标进行检测,探究野生植株与突变植株在离体处理模拟的逆境胁迫下各指标之间的变化。结果表明:突变植株与野生植株相比,在逆境胁迫下叶绿素含量下降幅度更小;逆境胁迫下,突变植株的MDA含量和可溶性蛋白含量更低;在非胁迫环境下,突变植株部分抗氧化酶活性更高。总体来看Zjsgr突变植株在离体处理下具有较好的持绿性,较强的抗氧化能力。综上所述,ZjSGR基因可能与植物抗逆性有关,为全面研究ZjSGR基因的功能奠定了基础,对选育结缕草滞绿、抗逆品种具有重要作用。

Abstract:Zoysia japonica is a perennial herb of the grass family,which is suitable for sports field turf planting,etc. However,its short green period in the north of China limits its use. Therefore,major breeding objectives of Zoysia japonica are longer green period and better resistance. The discovery of the STAY-GREEN gene has provided new ways to improve green retention. In this paper,we examined physiological indicators of wild type plants and ZjSGR mutant plants,including chlorophyll content,MDA content,soluble protein content and antioxidant enzyme activity,and found that the wild plants and the mutant plants showed different degrees of changes in each index under different in vitro treatments simulated by the stress of adversity. The results showed that compared with wild plants,the chlorophyll content decreased less in the mutant plants under stress;the MDA content and soluble protein content were lower in the mutant plants under stress;and some of the antioxidant enzyme activities were higher in the mutant plants under non-stress environment. Overall,Zjsgr mutant plants had better green retention and stronger resistance under stressed conditions. This paper demonstrates that the ZjSGR gene may play a role in Zoysia plant in response to stress,and it is important for the selection and breeding of stay green trait varieties of Zoysia grass.
