普通野生稻褐飞虱抗性遗传纯合研究

摘    要:目的]研究普通野生稻褐飞虱抗性遗传的稳定性,筛选出抗性稳定的野生稻种质,为水稻抗褐飞虱育种提供抗源.方法]对从46个原生地采集获得的1591份野生稻植株进行褐飞虱抗性分析,并选择具有抗性的野生稻材料通过套袋自交或花药培养分析其后代抗性遗传纯合表现.结果]收集的野生稻材料主要为杂合体,其自交一代的生长习性和芒性等发生明显分离.在1591份普通野生稻中,仅有30份材料对褐飞虱具有抗性,其抗性等级为3~5级,大部分为5级.在11份抗性材料的自交后代中,Z1~Z3均存在褐飞虱抗性分离,部分材料在Z4抗性表现稳定;有5份材料抗性从3级提高到1~3级,有4份材料抗性从5级提高到1~3级,有两份材料抗性从5级提高到3级.普通野生稻材料2174花药培养结果表明,6000枚花药经离体培养、诱导,可获得125块独立起源的愈伤组织,分化出8丛双倍体绿苗和两丛单倍体绿苗,愈伤组织诱导率仅为2.0%,绿苗分化率为6.4%.8个独立起源的花培后代中有6个株系抗性等级为5级,有两个抗性等级达到抗的水平(3级),未发现具有1级高抗水平的植株.结论]普通野生稻通过套袋自交和多代抗虫鉴定,可以获得稳定的高抗抗源,可以明显提高普通野生稻的褐飞虱抗性水平.花药培养有利于加速野生稻抗性遗传纯合,缩短后代筛选时间.

Abstract:Objective]The heredity stability of Oryza rufipogon resistance to brown plant hopper was studied to screen wild rice germplasm with stable resistance for providing sources in breeding resistant rice varieties.Method]Total 1591 accessions of Oryza rufipogon,collected from 46 proterozoic growth locations of Guangxi,were screened for brown plant hopper resistance and the genetic homozygosis in their offsprings was detected using bag-selfing or anther culture methods.Result]The results showed that most of Oryza rufipogon germplasm accessions were heterozygotic and the growth habit and awn of their self F1 generation showed separation.Among 1591 germplasm accessions,only 30 showed 3r d-5t h grades of resistance to brown plant hopper,most of them showed 5t h grade.Among self offsprings of 11 resistant accessions,the Z1-Z3 showed separation of resistant genes,while some accessions showed stable resistance in Z4.Out of 11 accessions,resistance of 5 accessions increased from 3r d to 1s t-3t h grades,4 accessions from 5t h to 1s t-3r d grades and 2 accessions from 5t h to 3r d grade.The anther culture of 2174 material showed induction of callus in 125 accessions from 6000 anthers.They were differentiated in 8 clusters of diploid and 2 clusters of haploid with 2.0% induction rate and 6.4% differentiation rate.Out of 8 offspring of anther culture,6 showed 5t h grade of resistance to brown plant hopper and 2 plants showed 3r d grade resistance.Conclusion]The stable highly-resistant resources could be obtained and resistance level of Oryza rufipogon could be enhanced through bag-selfing and multi-generation identification of resistance to brown plant hopper.Anther culture is beneficial to promote the genetic homozygosis of Oryza rufipogon resistance to brown plant hopper and shorten homozygosis time.
