摘    要:【目的】建立番茄水提物中总皂苷含量测定的方法,为进一步研究番茄皂苷奠定基础。【方法】成熟樱桃小番茄经打浆酶解后,所得清液经D-101大孔树脂吸附制得粗皂苷,以番茄皂苷A为标准品,以香草醛—冰醋酸+高氯酸体系作显色剂,于550 nm下测定总皂苷的含量。【结果】标准曲线在0.052~0.312 mg范围内线性关系良好(R2=0.9958);平均加样回收率为99.53%,RSD=0.52%(n=6);对5批次粗皂苷粉末中的总皂苷进行测定,其平均含量为(46.75±2.09)%。【结论】大孔吸附树脂—比色法操作简便、快速、准确,适用于测定番茄水提物中的总皂苷。

Abstract:Objective]A method for determining total saponin content in aqueous extract of tomatoes was developed to facilitate studies on tomato saponins. Method]After zymohydrolysis, tomato slurry obtained by smashing was subjected to D-101 macroporous resin to get crude saponins. Esculeoside A was used as reference and vanillin-perchloric acid was applied as chromogenic reagent to determine total saponin content at 550 nm. Result]The standard curve of esculeoside A showed good linearity in the range of 0.052-0.312 mg with R2=0.9958, and the average recovery of added sample was 99.53% with 0.52%(n=6) RSD. Total saponin content obtained in five batches of crude saponin extraction was found to be 46.75±2.09%. Conclusion]The method developed in the present study was rapid, accurate and easy to be used,therefore would be good for determination of total sapnion content in water extract of tomatoes.
