越桔工厂化育苗研究

摘    要: 本项研究以半高丛越桔‘Northsky’、‘Northblue’,矮丛越桔‘Blomidon’、‘Brunswick’4个栽培品种为试材,进行试管苗繁殖。用经过改良的WPM培养基加0.5—1mgL#+(-1)玉米素大量增殖幼枝。幼枝试管外扦插,用IBA1000、2000、3000ppm快速浸蘸法和2、5、10ppm慢速浸润法处理均能顺利生根;用塑料盘盛装腐苔藓基质,扣塑膜小棚保湿的简易设备进行试管外扦插,生根率达90%以上。研究中创造了塑料小盒冬季冷贮幼枝、透明塑料盒滤纸卷扦插吊挂空中催根等方法,更便于苗木工厂化生产。同时发现了越桔试管苗有因饥饿和冷凉而被迫的绿色休眠习性,可做为研究大量冬贮幼苗的理论依据。本文根据越桔特性提出在生态最适区建立工厂化育苗的配套培育圃。最后论证了越桔组织培养苗可以用于生产性栽培。

STUDY ON FACTORY-TYPE PROPAGATION OF BLUEBERRY

Abstract:Micropropagation of 2 cultivars of half-highbush blueberry 'Northsky' and 'Northblue', and 2 cultivars of lowbush blueberry 'Blomidon' and 'Brunswick' were studied. Multiple shoot proliferation were induced on modified WPM medium with 0.5-1mgl~(-1) zeatin. Shoots were rooting successfully ex vitro both by dipping rapidly with IBA at 1000, 2000, 3000ppm and moistening for long with IBA at 2, 5, 10ppm. The rooting percentage of shoot cuttings were more than 90% under ex vitro conditions with peat moss medium in plastic shallow cases arched by a lower plastic film tent to keep a high humidity. Two inventive methods were established: (1) using plastic boxes for cool storage of the proliferated shoots in winter; (2) hanging the clear plastic boxes containing the proliferated shoots with their base wrapped in a moist filter paper tape for rooting. Both are more suitable for the factory-type propagation of blueberry young plants. It was found that the normal defoliating dormancy were not present in blueberry young plants by tissue culture, but the forced green dormancy arose from starving or cooling. This discovery may be as a theoretical basis for the study on winter storage of mass young plants by tissue culture. A suggestion on establishing complete set nursery in the most suitable ecological locations for blueberry growing has been put forward.At the end of this paper, it is discussed that the tissue culture plants of blueberry are available for commercial growing.
