水稻黑条矮缩病毒在不同抗性玉米自交系叶片内的积累研究

摘    要:应用玉米粗缩病病情严重度分级标准,对已知抗性水平的抗病、中抗和感病自交系材料逐株进行病情严重度分级调查。采集不同级别病株的玉米新叶,用间接酶联免疫吸附法(Indirect enzyme-linked immunosorbentassay,ID-ELISA)检测病株叶片中水稻黑条矮缩病毒(Rice black-streaked dwarf fijivirus,RBSDV)。采用SPSS软件对O.D405光密度值进行差异显著性分析。结果显示:同一抗性水平、不同级别的玉米自交系病株叶片内的RBSDV浓度差异显著(F=237.499,P<0.01),且随病株病级加大,病毒浓度升高;而相同病级的抗病、感病和中抗玉米自交系叶片内的RBSDV浓度无显著差异(F=0.775,P=0.598);抗病自交系病株叶片中的病毒总浓度显著低于感病自交系材料。试验结果表明:玉米粗缩病病株叶片内的病毒浓度与生物学症状呈正相关,病情越重,病毒浓度越高。RBSDV一旦侵入寄主,在抗病、感病和中抗的自交系植株体内均可增殖,并运转到新生叶片。此结论为从分子水平上进一步揭示玉米抗粗缩病的机理奠定了基础。

Abstract:According to the height of the plants and symptoms on leaves,a criterion of 0-4 to examine the disease severity was reported previously.In this study,the criterion was adopted to grade the disease severity of the tested corn inbred lines with different resistance to the disease during the milky growth stage.Indirect enzyme-linked immunosorbent assay(ID-ELISA) was applied to detect rice black-streaked dwarf fijivirus(RBSDV) in newly sprout leaves.Variance analysis of the O.D405 values showed that there were significant differences on RBSDV concentrations among the different graded plants of the same inbred line.RBSDV concentrations were higher as the disease grading went up from 1 to 4 grades(F=237.499,P<0.01).While in same disease grade of the resistant and susceptible inbred lines,there was no obvious difference on RBSDV concentration(F=0.775,P=0.598).This indicated that RBSDV concentration in leaves was positively related to the symptoms of whole plants;RBSDV could multiply in corn inbred lines with different resistance and move to the new growth leaves.
