摘    要:为评价不同家系和不同生长阶段杉木种子酚类物质含量差异,采用超声波提取法提取杉木涩粒和健籽的酚类物质,分别考察提取次数、提取时间、乙醇浓度、提取温度和液料比对杉木涩粒和健籽多酚提取效果的影响,以单因素试验的结果为依据,设计L9(34)正交试验,进一步对涩粒和健籽的多酚提取工艺进行优化。结果表明:涩粒的最佳提取工艺是以50%乙醇为提取剂,液料比为60∶1(m L∶g),在提取温度为50℃条件下处理15 min,提取1次,涩粒的多酚提取量达到(383.220±1.233)mg·g-1。健籽在乙醇浓度为100%,提取温度为50℃、液料比为25∶1(m L∶g)、提取时间为25 min条件下提取1次,其多酚提取量达到最高值,为(18.360±0.069)mg·g-1。验证实验表明,优化后的条件参数稳定可靠。

Abstract:In order to evaluate the variation of polyphenols in the seeds of different families of Chinese fir at different growth phases, and offer technical support for elucidating the underlying mechanisms of the abortive seeds formation in Chinese fir. An ultrasonic-assisted extraction method was applied to the effective extraction of polyphenols from the abortive and healthy seeds of Chinese fir. The effects of extraction times, extraction duration, ethanol concentration, extraction temperature and solvent-to-sample ratio on the yield of polyphenols from the seeds of Chinese fir were observed, respectively. Based on the results of single-factor experiments, an orthogonal design was employed to further optimize the extraction process. The results showed that the optimal extraction conditions for abortive seeds were 50% ethanol concentration, the solvent-to-sample ratio was 60 ∶ 1, and extraction for one time at 50 ℃ for 15 min. Under these optimal conditions, the maximum yield of polyphenols from abortive seeds was (383.22±1.23) mg·g-1. At the same time, the optimal extraction conditions for healthy seeds were 100% ethanol concentration, the solvent-to-sample ratio was 25∶ 1, and extraction for one time at 50℃ for 25 min. Under these optimal conditions, the maximum yield of polyphenols from healthy seeds was (18.360±0.069) mg·g-1. Verification test showed that the optimized extraction parameters were stable and reliable.
