银丝竹不同叶色叶绿素合成及叶结构差异

摘    要:为探明银丝竹叶色差异的原因,对银丝竹不同叶色叶片的光合色素含量、 叶绿素合成前体相对含量及叶片显微和超微结构进行研究.结果表明:1)银丝竹不同叶色间的光合色素含量存在显著差异,其含量随着绿色叶片面积的下降而下降;2)银丝竹叶片出现白色性状的原因在于其叶绿素生物合成过程受阻,受阻点可能位于粪卟啉原Ⅲ至原卟啉原Ⅸ的转化过程中;3)两种叶色叶片表皮细胞无明显差异,但白色叶肉细胞中内容物明显缺失;4)绿色叶肉细胞中具有丰富而完整的叶绿体结构,而白色叶肉细胞中呈现出叶绿体膜结构模糊,无完整边界等形态,基粒类囊体片层不规则或呈空泡状,在多数细胞内可见较明显的嗜锇颗粒.本研究从生理层面初步解释了银丝竹叶色差异的原因,为进一步开展相关分子机理研究奠定了基础.

Abstract:In order to find the reason of leaf color mutation, the trial studied the content of photosynthetic pigment, level of chlorophyll synthesis precursor and microstructure and ultrastructure of different color leaves of Bambusa multiplex 'Silverstripe'. The results showed that: 1) there was a significant difference among the photosynthetic pigment content in different type leaves, and the content decreased with the decrease of green leaf area; 2) the reason for showing a white leaf was the chlorophyll biosynthesis block and the blocked points would be in the conversion process of Coprogen Ⅲ to Proto Ⅸ; 3) there was no significant different between the epidermis of 2 kinds leaves, whereas the content in the white mesophyll cells were absent; 4) compared with the intact chloroplast structure in green mesophyll cells, dysplastic chloroplasts could be observed in white one, including chloroplasts with swollen thylakoids and fragmented or no chloroplast membranes, and where the osmiophilic granules could be seen. This study preliminarily explained the reasons for the differences in the leaf color of B. multiplex 'Silverstripe', and laid the foundation for further research on the molecular mechanism.
