| 摘 要： | 古树名木是大自然经过漫长的历史变迁,留给人类的宝贵遗产,具有重要的科学、文化和经济价值.为了传承历史文化,留传历史园林植物的宝贵遗传种质资源,以北京颐和园西堤古柳嫩枝为外植体进行茎段组织培养试验研究.结果表明:取古柳树外植体带有侧芽的茎条切段进行组织培养,以MS为基本培养基,附加不同浓度的6-BA和NAA进行分化诱导,其中最适启动培养基为不添加任何激素的MS培养基(2.0%蔗糖);最适不定芽诱导培养基及激素配比为MS+6-BA0.8mg/L+NAA 0.2 mg/L+3.0%蔗糖;不定根最适诱导培养基为1/2MS+NAA0.2mg/L+1.5%蔗糖.

| Abstract: | Old trees are the precious legacy of the long history with important scientific,cultural and economic value.In order to inherit the historical culture and reserve the valuable plant genetic resources of the old national garden plants,tissue culture technique of the ancient willows grown on the west dyke from the Summer Palace in Beijing was studied.The result indicated that clustering plants could be gained by using stem with lateral bud as explants,taking MS as basal medium,supplement with 6-BA and NAA at ... |
