家兔精液冷冻保存的研究

摘    要:以家兔为试验动物,通过精液在4℃保存时间来筛选合适的基础液,并从精液解冻后的精子活率和活力2个指标筛选出最佳的平衡时间和抗冻剂(甘油)浓度.冷冻保护液中渗透性抗冻剂(甘油)的3种浓度为3%,4%和5%.4℃平衡时间分别为2,3,4h.将3种甘油浓度的冷冻保护液与3种平衡时间相配伍,共9组进行冷冻试验,筛选出稀释液为:Tris3.03g,柠檬酸1.30g,果糖1.50g,卵黄20%,青、链霉素各1×10^5U,双蒸水100mL.冷冻后结果表明,3%的甘油浓度平衡3h,解冻后精子活力在0.3以上,达到人工授精的要求.

Abstract:Take the Rabbit as experimental animal. Screening the appropriate foundation fluid through the seminal fluid in 4℃ retention time, and according to frozen-thawing sperm recovery and mobility rate of sperm screens the best equilibrium time and the antifreeze (glycerine) density. In freezing protection fluid permeable antifreeze (glycerine) three kinds of densities was 3%, 4% and 5%, 4℃ equilibrium times was 2,3 and 4 h, respectively. Combining three kinds of glycerine densities freezing protection fluid with three kinds of equilibrium time, 9 groups were studied. Screened the diluent is tris 3.03 g, citric acid 1.30 g,fructose 1.50 g,yelk 20%, penicillin 1×10^5U, streptomycin 1×10^5 U,D. W 100 mL. The freezing results showed that 3% glycerine density balance for 3h, frozen-thawing sperm mobility above 0.3. Achieved the artificial insemination the request.
