中国荷斯坦奶牛PPARGC1A基因内含子9的SNP与产奶性状的相关分析

摘    要:采用PCR产物直接测序的方法,对213头中国荷斯坦奶牛的PPARGC1A基因进行单核苷酸多态性分析,并研究不同基因型与产奶性状的相关关系。结果表明,PPARGC1A基因内含子9存在C>T SNP位点,可分为有CC、CT和TT这3种基因型,基因型频率分别为0.3568、0.6291和0.0141;该位点突变偏离Hardy-Weinberg平衡状态且中度多态;CC基因型与中国荷斯坦奶牛的乳脂肪率显著相关。PPARGC1A基因可以作为研究中国荷斯坦奶牛泌乳性状的候选基因。

Abstract:This study determined single nucleotide polymorphism(SNP) of PPARGC1A in 213 Chinese Holstein cattle by DNA sequencing and investigated the association between genotypes and milk production traits.Results showed there was one variation site(C>T) located at intron 9,we assigned CC,CT,TT as three genotypes for the studied gene and the genotype frequencies were 0.3568,0.6291,0.0141 respectively;the variable site deviated Hardy-Weinberg equilibrium and had intermediate polymorphism information contents;CC genotype had significant association with milk fat percentage.Therefore,PPARGC1A gene could be regarded as a candidate gene effecting milk traits.
