摘    要:目的]筛选白芨原球茎诱导和增殖的最适培养基,建立白芨原球茎高效增殖体系,为白芨种苗工厂化组培生产提供技术支持.方法]以白芨种子为外植体,筛选适宜其无菌萌发的灭菌方法;以MS或1/2MS为基本培养基,添加不同浓度的植物生长物质和附加物,通过单因素试验,确定最佳光照和温度条件,筛选最适合其原球茎诱导的培养基;通过正交试验,筛选最适合其增殖的培养基.结果]采用75%乙醇处理白芨蒴果10 min,无菌萌发率达90.0%,且种子萌发不受影响.在原球茎诱导过程中,30.0 μmol/m2·s光照强度和25 ℃温度环境有利于原球茎诱导;最适合原球茎诱导的培养基为1/2MS+1.0 mg/L 6-BA+0.1 mg/L NAA.在原球茎增殖过程中,添加土豆泥、椰子汁、香蕉泥等附加物对原球茎增殖有益,其中添加50.0 g/L土豆泥的增殖效果最佳.正交试验结果表明,原球茎增殖的最佳培养基为MS+0.5 mg/L 6-BA+0.2 mg/L NAA+50.0 g/L土豆泥,增殖倍数达5.270.结论]建立的白芨原球茎高效增殖体系可在白芨种苗工厂化组培生产中推广应用.

Abstract:Objective]The optimal medium for Bletilla striata(Thumb.)Reichb.f.protocorm induction and prolifera-tion were screened and effective protocorm proliferation system of B.striata was establishand to provide technical support for industrialized production of tissue culture seedlings of B. striata.Method]B. striata seeds were used as explant to select the proper methods of aspetic germination.Taking MS or 1/2MS as the basic medium,and adding plant growth sub-stance with different concentrations and additives to determine the optimum light and temperature conditions by single factor test as well as select the most suitable medium for its protocorm induction.Through orthogonal test,the most suitable medium for its proliferation was screened.Result]After treating B.striata capsule by 75% ethyl alcohol for 10 min,the aspetic germination rate could reach 90.0% and the seed germination was not affected.In the process of proto-corm induction,30.0μmol/m2·s illumination intensity and 25℃temperature benefited the germination of protocorm.The optimal medium for protocorm induction was 1/2MS medium+1.0 mg/L 6-BA+0.1 mg/L NAA.During the proliferation of protocorm,the addition of mashed potato,coconut juice and banana mud was beneficial to the proliferation of protocorm, and 50.0 g/L mashed potato had the best effects. The orthogonal test showed that the optimum medium for protocorm proliferation was MS+0.5 mg/L 6-BA+0.2 mg/L NAA+50.0 g/L mashed potato,and the proliferation mutiple reached 5.270.Conclusion]The established effective B.striata protocorm proliferation system can be widely used in the factory in the production of tissue culture seedlings of B.striata.
