摘    要:目的]研究NH4+引起炎症反应的机制。方法]利用倒置显微镜观察不同盐离子引起单核癌细胞THP-1形态的变化,乳酸脱氢酶(LDH)试剂盒检测细胞培养上清中LDH,台盼蓝染色检测细胞形态和存活情况,Western blot检测炎症反应相关细胞因子的产生和分泌。结果]通过体外研究发现NH4+能够激活caspase-1和下游的IL-1β,IL-1β是炎症反应的重要细胞因子;K+能够抑制NH4+引起的caspase-1和IL-1β的激活。结论]Na+、SO24-、Cl-都不引起炎症反应,对THP-1细胞没有影响;NH4+能够引起单核细胞的炎症反应,并且可以被K+抑制,为氨基酸代谢紊乱引起的疾病的治疗提供了一定的依据。

Effect of Different Ionic Salts on Monocyte Inflammatory Reaction

Abstract:Objective]The aim was to study the mechanism of inflammatory response caused by NH4+.Method]By using inverted microscope,the variation of monocyte THP-1 morphology caused by different ionic salts were detected,moreover,based on LDH kit to detect LDH in cell culture supernatant,what's more,through trypan blue staining to detect cell morphology and survival,and the production and secretion of inflammatory cytokine were detected by Western blot.Result]In vitro study,we found that NH4+ could activate caspase-1 and downstream cytokine IL-1β,IL-1β was an important inflammatory cytokine;K+ can inhibit caspase-1 and IL-1β activation.Conclusion]Na+,SO2-4,Cl-irons did not cause inflammation of the THP-1 cell;NH4+ can cause monocyte cells inflammation,and the inflammation can be inhibited by K+ ion,which provided certain basis for disease treatment caused by amino acid metabolism disorders.
