Abstract:Genomic in situ hybridization (GISH) and acid polyacrylamide gel electrophoresis (A PAGE) of gliadin were used to identify the alien chromatin in a novel germplasm (A 3) derived from a wheat Thinopyrum ponticum hybrid. Using genomic DNA of "Chinese Spring"( Triticum aestivum L. ABD) as probe, genomic DNA of Pseudoroegneria stipifolia (one possible genome donor of Th. ponticum, its genome is St) as block, one pair of Robertsonian translocation chromosomes were detected via GISH. An 18S 5.8S 26S rDNA locus was located on the alien arm by FISH of p Ta 71, a rDNA sequence isolated from wheat. Further analysis of GISH with genomic DNA probes of the three diploid progenitors of T. aestivum respectively indicated that the translocated chromosomes were concerning the B genome chromosomes. The result of A PAGE of gliadin showed that A 3 conveyed the 1RS specific gliadin marker Gld B3. Using rye ( Secale cereale L.R) genomic DNA as probe and common wheat genomic DNA as block, it was then proved that A 3 was a truly 1BL/1RS translocation line by GISH. The risk for making false explanation on GISH signals caused by cross hybridization between genomes was discussed. The results indicate that we must be prudent in explanation of the GISH signals and identification of alien chromosome. It is easy to draw a false conclusion if only a single technique is adopted. Multiple methods should be used to confirm the results and check with each other.
