动植物总核酸提取试验

摘    要:依据最简单的核酸提取3步骤:细胞裂解、去除杂质、沉淀核酸的原理设计了一种普适性的动植物核酸提取方法(简称PS法),并对包括裸子植物门的银杏纲、苏铁纲、松柏纲,被子植物门的双子叶植物纲和单子叶植物纲的20种植物材料以及动物界的5纲5种材料进行了提取试验.结果表明,用PS方法提取动植物总核酸均能取得较理想的提取效果.所提取的总核酸分别用RNase和DNase进行消化,可分离纯化出高纯度的DNA和RNA.

Abstract:According to simple three steps for nucleic acid extraction,in cluding cells rupturing,impurities removing and nucleic acid precipitation a new nucleic acid extraction method called PS method was worked out.Better results were obtained on the basis of extractions from 20 species of plants including Ginkgopsida,Cycadinae,Coniferae,dicots and monocotyledon of angiosperms,and 5 species of animals of 5 classes.High purity of DNA and RNA were obtained after digestion with DNase and RNase respectively.The extracted DNA and RNA were excellent for RT-PCR and SSR amplifications.The experimental results showed that the PS method was a broad-suitable method for nucleic acid extraction from plants and animals.
