牦牛应激型 HSPA1AHSPA2基因的克隆及序列分析

摘    要:为了解牦牛应激型 HSPA1AHSPA2基因的特点,根据GenBank已公布的牛应激型 HSPA1AHSPA2基因序列设计4对引物,每个基因分两段扩增,测序并拼接,首次克隆牦牛应激型 HSPA1AHSPA2基因。序列分析表明,扩增到的牦牛应激型 HSPA1A基因全长2 134 bp,开放阅读框全长1 926 bp,编码641个氨基酸,分子质量为70.26 ku; HSPA2基因全长1 911 bp,开放阅读框全长1 911 bp,编码636个氨基酸,分子质量为69.85 ku。将 HSPA1AHSPA2基因开放阅读框编码的氨基酸序列进行ScanProsite分析,均得到3个HSP70蛋白家族标记。核苷酸序列同源性分析表明, HSPA1AHSPA2基因具有较高的保守性,牦牛与牛的 HSPA1AHSPA2基因核苷酸序列同源性最高,分别为99.8%和99.1%。遗传进化关系分析表明,牦牛与牛的应激型 HSPA1AHSPA2基因亲缘关系最近。

Cloning and Sequence Analysis of Stress Type HSPA1A and HSPA2 Genes from Yak

Abstract:To understand the character of stress type HSPA1A and HSPA2 genes of yak, four pairs of primers for HSPA1A and HSPA2 genes of yak was designed according to the published HSPA1A and HSPA2 genes sequence of Bos taurus in GenBank. Each gene was divided into two sections for amplification, sequencing and splicing, and succeeded in cloning stress type HSPA1A and HSPA2 genes of yak for the first time. The results of sequence analysis showed that the length of the cloning HSPA1A cDNA is 2 134 bp, containing 1 926 bp open reading frame (ORF) , encoding a peptide of 641 amino acids with molecular mass of 70.26 ku; the length of the cloning HSPA2 cDNA is 1 911 bp, containing 1 911 bp ORF and encoding a peptide of 636 amino acids (69.85 ku). Analysis of amino acid sequences with ScanProsite, three signature sequences of HSP70 family were found for each gene. Sequence homology analysis indicated that HSPA1A and HSPA2 genes are highly conservative, and the highest homology, 99.8% and 99.1% respectively, was detected between yak and Bos taurus. The genetic evolution relationship analysis showed that stress type HSPA1A and HSPA2 genes of yak and Bos taurus have the closest genetic relationships.
