生物反应器在金线莲扩繁中的应用

摘    要:目的]研究在自动化低成本的生物反应器下内大量扩繁金线莲的简单方法。方法]把金线莲丛状芽培养在不同环境条件的3L球形生物反应器中,加入花宝(20-20-20)1g/L+(6.5-4.5-19)1g/L培养基,添加蛋白胨2g/L、蔗糖3%和颗粒活性炭0.5g/L,比较b凝胶培养基与不同条件下生物反应器(浸没式、接触式和潮汐式)培养的金线莲丛生芽的生长情况。结果]植株生长(干重、茎高、叶数、根数和根长)在接触式生物反应器中结果相对较好。当外植体数目在60~90个、通气量在0.06vvm时测试结果(鲜重、茎长、叶数、叶面积和根数)最好,再生苗拥有良好的根系且在温室容易驯化成功。结论]利用生物反应器扩繁金线莲是一种新的有效途径。

Abstract:Objective] This study was to develop an autonomic and low cost bioreactor system for the proliferation of Anoectochilus formosanus.Method]The mulitple shoots of A.formosanus were cultured in three-liter balloon type bubble bioreactors under different conditions by adding 1.0 g/L the ready-to-use media Hyponex(20-20-20)and 1.0 g/L ready-to-use Hyponex(6.5-4.5-19),2.0 g/L peptone,30 g/L sucrose and 0.5 g/L active carbon granules,and various gelatin media and culture methods(continuous immersion with or without bracing net and temporary immersion)were also compared.Result]In the bioreactor of continuous immersion with bracing net,A.formosanus plantlets grew best in dry weight,stem length,leaf number,root number and root length under other parameters of 60-90 explants,0.06 vvm aeration volume.The plantlets regenerated had well developed root system and were easily acclimatized in greenhouse.Conclusion]The system developed in the present study could be an effective approach to proliferate A.formosanus.
