茉莉酸甲酯对地黄毛状根次生代谢的影响

摘    要:以地黄毛状根为试材,茉莉酸甲酯作为诱导子,采用HPLC法测定梓醇与毛蕊花糖苷含量,采用紫外分光光度法测定总环烯醚萜苷含量,研究了地黄毛状根在茉莉酸甲酯诱导下的次生代谢产物积累规律,以期提高地黄毛状根中有效成分的产量。结果表明:地黄毛状根中未能检测到梓醇;地黄毛状根中毛蕊花糖苷含量在茉莉酸甲酯诱导后的第2天达到最大值,约为对照组的1.41倍。地黄毛状根中总环烯醚萜苷含量在茉莉酸甲酯诱导后变化不显著。在茉莉酸甲酯诱导初期,毛蕊花糖苷的合成显著增加,随着诱导时间的延长,较高的毛蕊花糖苷浓度也可能负反馈抑制其合成途径中关键酶的活性,从而使其在诱导后期含量下降。此外,50μmol·L^-1的茉莉酸甲酯对地黄毛状根总环烯醚萜苷积累的诱导作用可能不明显。

Effects of Methyl Jasmonate on Secondary Metabolite Production in Rehmannia glutinosa Libosch. Hairy Root Culture

Abstract:Taking hairy roots of Rehmannia glutinosa Libosch as materials,methyl jasmonate was used as the elicitor.The contents of catalpol and verbascoside were determined by HPLC method.The content of total iridoids was determined by UV spectrophotometry.The accumulation of secondary metabolites of hairy roots induced by methyl jasmonate were explored,in order to increase the accumulation of secondary metabolites in hairy roots of R.glutinosa.The results showed that catalpol was not detected in the hairy roots of R.glutinosa.The content of verbascoside in the hairy roots of R.glutinosa reached its maximum on the second day after the induction of methyl jasmonate,which was about 1.41 times that of the control group.The content of total iridoids did not change significantly after induction with methyl jasmonate.In the early period of induced by methyl jasmonate,the synthesis of verbascoside significantly increased.However,with the extension of the induction time,the higher glucosinolate concentration might also inhibit the activity of key enzymes of verbascoside biosynthesis pathway,thereby reducing its content in the late induction period.Meanwhile,50 μmol·L^-1 MeJA might have no effects on accumulation of iridoids according to available data.
