平贝母愈伤组织和不定芽诱导研究

摘    要:目的应用组织培养技术对平贝母进行离体培养,从而得到愈伤组织和不定芽。方法以鳞茎为材料诱导愈伤组织与不定芽,采用正交实验设计确定最佳的激素种类与浓度。结果适当的激素组合及配比可诱导出愈伤组织和不定芽,最高的诱导率可分别达到14%以上、86.7%。结论愈伤组织最佳诱导培养基为MS+2,4-D 1.0mg/L+6-BA 0.9mg/L+NAA 0.5mg/L;最佳继代培养基为MS+2,4-D0.5mg/L+6-BA 0.3mg/L;不定芽最佳诱导培养基为MS+KT 0.8mg/L+NAA 0.3mg/L。

Studies on Callus and Adventitious Bud Induction of Fritillaria ussurionsis Maxim

Abstract:Objective To apply the tissue culture technology for the callus and adventitious buds of Fritillaria ussurionsis Maxim in vitro.Methods The optimum hormone types and concentration for the callus and adventitious bud from the bulbs were determined with the orthogonal experiment.Results The optimum hormone types and concentration could induce callus and adventitious bud,the highest inducing rates are more than 14%,86.7% respectively.Conclusion The optimum callus induced media: MS+2,4-D 1.0mg/L+6-BA 0.9mg/L+NAA 0.5mg/L,the successive transfer media: MS+2,4-D 0.5mg/L+6-BA 0.3mg/L;the adventitious bud media: MS+KT 0.8mg/L+NAA 0.3mg/L.
