摘    要:以玫瑰(Rosa rugosa)叶片为材料,利用改良的CTAB法,提取到了高质量的玫瑰叶片DNA,并建立了37个玫瑰品种和5个蔷薇品种的AFLP银染反应体系,首次在玫瑰DNA提取、酶切连接、预扩增、选择性扩增等实验过程取得了成功,得到了清晰的玫瑰品种的AFLP指纹图谱,为玫瑰基因库的建立、优良品种选育以及亲缘关系的系列研究奠定了理论基础.

Abstract:Extraction of DNA with phenol-cholorofore method from Rosa rugosa leaves was dealt with and AFLP finger printing in 37 R.rugosa cultures and 5 Rosa davurica were established.Clear and high resolution DNA finger prints were obtained,which mena success in DNA extraction,restriction-ligasereaction,pre-amplification and staining techniques.A foundation for the establishment of gene pool,selection and breeding of eminent cultivars and geneic relaionshpi in R.rugosa was laid.
