油菜雄性不育基因工程植株的建立及鉴定

摘    要:用含有抗除草剂溴苯腈与雄性不育嵌合基因、抗除草剂溴苯腈与育性恢复嵌合基因转化油菜下胚轴组织,分化再生获得转基因雄性不育植株和育性恢复植株。用除草剂溴苯腈对转基因雄性不育植株和育性恢复植株进行抗性鉴定,抗性可达10-3mol/L。再以抗溴苯腈基因为探针分别对其进行Southern杂交分析,杂交结果均为阳性。对两种植株花器解剖表明,雄性不育植株的花丝明显比花柱短,花粉少,育性恢复植株与正常植株相同

Establishment and Detection of Transgenic Male Sterile Plants in Brassica napus
Abstract:The method of breeding Brassica male sterile and fertile restorer plants using genetic engineering techniques was researched and these plants were detected.Transgenic male sterile and fertile restorer plants in Brassica were obtained by transforming the chimeric genes of anti Basta gene and male sterile gene and the chimeric genes of anti Basta gene and fertile restorer gene to hypocotyl explants of Brassica.These transgenic plants were detected using herbicide Basta,the conc.of resistance reached to 10 3 mol/L.The result of Southern blot of these plants was positive.The flowers of transgenic plants were dissected,filaments of transgenic male sterile plants were much longer than their styles and their pollens were lessened,however,the transgenic fertile restorer plants were normal as control plants.
