几个转基因植株后代的遗传分析

摘    要:对用农杆菌介导的遗传转化法获得的9个独立转化子进行了遗传分析。结果显示:除转基因植株R1Ab-8外,其他8个转基因植株都出现选择标记基因bar与目标基因cry1Ab进行共整合。单拷贝转基因植株中,除R1Ab-6植株外,其余4个转基因植株均表现3∶1的孟德尔式的遗传规律。双拷贝转基因植株R1Ab-3出现15∶1的分离比,说明两个拷贝的外源基因整合到受体基因组的不同染色体;双拷贝转基因植株R1Ab-7出现3∶1的分离规律,说明两个拷贝的外源基因整合到受体基因组的同一染色体。同时,还发现R0代表现抗性且出现孟德尔分离比的单拷贝转基因植株的纯合体的杂种F1代也能表现抗虫性,表明,利用转基因培育作物新品种时,应选择单拷贝、符合孟德尔遗传且表现目标性状的转基因植株作母本。

Abstract:9 independent transformants got by genetic transformation system mediated with agrobacterium were conducted for inheritance analysis.The results showed that except for transgenic plant R(1Ab)-8,the other 8 transgenic plants all co-integrated selective marked gene bar with target gene cry1Ab.Among the single-copy transgenic plants,except for plant R(1Ab)-6,the other 4 transgenic plants all showed Mendelian inheritance law of 3∶1.The double-copy transgenic plant R(1Ab)-3 showed separation ratio of 15∶1,indicating that 2 copied exogenous genes was integrated into different chromosomes in acceptor genome.The double-copy transgenic plant R(1Ab)-7 showed separation law of 3∶1,indicating that copied exogenous genes was integrated into the same chromosomes in acceptor genome.Besides,it was also discovered that R0 generation showed resistance and the hybrid F1 of homozygote in single-copy transgenic plants Mendelian separation ratio appeared could show pest-resistance too.These showed that in breeding new crop variety by transgenic technology,the single-copy transgenic plant according with Mendelian inheritance law and showing target characters should be chosen as female parent.
