| 摘 要： | 目的]研究猪圆环病毒2型ORF2的基因克隆及其在E.coli中的融合表达。方法]参照猪圆环病毒2型ORF2序列设计引物,应用PCR技术扩增出猪Ⅱ型圆环病毒河南株ORF2的部分基因。将其同IL-2的全基因一起连接PBV220载体并转化到E.coliJM109。阳性菌用温度诱导表达,表达产物经SDS-PAGE和Western-blot分析。结果]结果表明,ORF2基因同IL-2的全基因在E.coli中得到了高效表达,表达的融合蛋白分子量为44 kD,可以被猪PCV2阳性血清特异性识别。结论]该研究为PCV2的检测和疫苗的研制以及综合防制奠定了基础。

| Abstract: | Specific primers were designed according to the sequences of ORF2 gene of porcine circovirus 2(PCV2) isolated from Henan.ORF2 gene was obtained by PCR from PCV2 of Henan.Together with the IL-2 gene,the DNA fragment of ORF2 gene was cloned into prokaryotic expression vector PBV220.SDS-PAGE and Western blot assays indicated that the fusion proteins with molecular weight of 44 kD were efficiently expressed in E.coli JM109,and the polyclonal antibody against PCV2 could recognize this protein.This study has laid a foundation for PCV2 detection and vaccine development as well as for its comprehensive control. |
