基于SSR和SRAP标记苦瓜种质遗传多样性分析

摘    要:种质资源遗传多样性是苦瓜遗传改良的材料基础。用SSR、SRAP标记对46份苦瓜种质遗传多样性进行分析及评价,结果表明,从26对SSR和196对SRAP引物中分别筛选获得14和33对多态性引物,并分别扩增出70和637条谱带,其中多态性条带分别为60和90个。利用软件进行聚类分析,46份供试苦瓜种质的遗传相似系数在0.61~0.88之间,取阈值0.656可将46份苦瓜分为4大类群。依据供试材料的亲缘关系,可以为亲本选配和杂种优势利用提供理论基础。

Abstract:Germplasm resource is the material basis for genetic improvement of Momordica charantia.Genetic diversity of 46 M.charantia germplasms was analyzed and evaluated with 14 pairs of SSR and 33 pairs of SRAP primers.The results showed that 60 of the 70 amplicons obtained by SSR primers and 90 of the 637 amplicons obtained by SRAP primers were polymorphic.Cluster analysis was conducted using software.It was concluded that the genetic similarity coefficient among 46 M.charantia germplasms ranged from 0.61 to 0.88,at the threshold of 0.656 where all the tested accessions were divided into 4 groups,which provided a theoretical basis for parents selection and utilization of heterosis based on genetic relationship of the test materials.
