甘薯抗蔓割病的生理机制Ⅱ.甘薯抗蔓割病与多酚氧化酶及其同功酶之间的关系

摘    要:对4个不同抗性的甘薯品种经苗茎剪口接种蔓割病菌后,测定叶片多酚氧化酶(PPO)活力及其同功酶,结果表明:接种第2dPPO活力上升,PPO活力病/健比值大于1,抗、感病品种之间差异不明显,第4、8d抗、感病品种PPO活力都下降,PPO活力病/健比值小于1,并且感病品种比值小于抗病品种;在接种的第8d叶柄PPO同功酶带没有增加,抗病品种扫描表面活力变化不大,而感病品种PPO同工酶酶谱颜色加深,条带不清,扫描表面活力明显降低.上述结果可作为鉴别品种抗性的生理指标

Abstract:The activity and isozyme of polyphenol oxidase( PPO) were assayed in the leaves of 4 cultivars of sweet potato with different resistance to fusarium wilt after inoculation with fusarium wilt fungus, which indicated that PPO activity increased, and the ratio of PPO activity in diseased plant to that in the healthy plant was greater than one, but there was no evident difference between the resistant cultivars and the sensitive ones on the second day of inoculation. On the fourth and eighth day, PPO activities decreased, the ratio of the diseased plant to the healthy plant was lower than one, and the ratio in sensitive cultivars was lower than that in the resistant ones. The PPO isozyme bands didn't increased in petiole on the eighth day of inoculation, and the scanning apparent activity changed little in the resistant cultivars. However, the PPO isozyme band patterns became unclear resulted from the deepening zymograms, the scanning apparent activity evidently decreased in the sensitive cultivars. The results suggested that PPO could be used as a physiological index to identify the resistance to fusarium wilt in sweet potato.
