黑番鸭VIPR-1基因单核苷酸多态性与就巢性状的关联性分析

摘    要:VIPR\|1基因是研究家禽就巢性状的重要候选基因,研究选用处在产蛋期、就巢期的黑番鸭为研究对象,通过PCR\|RFLP和测序的方法,对VIPR\|1基因内含子12进行多态性检测,并分析其与就巢性状之间的关系。结果表明:VIPR\|1基因内含子12存在一个AfaⅠ酶切位点,该位点在内含子12的902 bp处发生了C→T的碱基突变,产生了CC,CT和TT三种基因型。通过对产蛋黑番鸭与就巢黑番鸭个体的研究,发现所测产蛋黑番鸭基因型的分布情况为CC(32),TT(40)和CT(42),就巢黑番鸭基因型的分布为CC(18),TT(40)和CT(18),经卡方独立性检验,产蛋黑番鸭与就巢黑番鸭两者之间基因型的分布差异显著(P<005)。表明黑番鸭VIPR\|1基因第12内含子C902T突变与黑番鸭的就巢性状显著相关。

Abstract:As a critical candidate gene of poultry broodiness, intron 12 of the vasoactive intestinal peptide\|1(VIPR\|1) gene of black muscovy ducks was analyzed by PCR\|RFLP. The results indicated that there was a site of AfaⅠ digestion within intron 12 of VIPR\|1 gene. An C→T mutation at the site 902 bp of this gene was detected by DNA sequencing which produced three sorts of genotypes CC, TT and CT, and showed genotype frequencies of CC(18), TT(40) and CT(18) in brooding black muscovy ducks, rather than CC(32), TT(40) and CT(42) in laying black muscovy ducks. By chi\|square test, there were significantly differences (P<005) between broodiness black muscovy ducks and laying black muscovy ducks in genotypes. This indicated that C902T mutation in intron 12 of VIPR\|1 gene was significantly associated with broodiness of black muscovy ducks.
