苹果茎痘病毒RT-PCR检测及分子变异分析

摘    要:利用RT-PCR技术对采自6个苹果品种和7个梨品种的33个样本中的苹果茎痘病毒(Applestem pitting virus,ASPV)进行了检测,ASPV感染率为63%(19/33)。对6个ASPV中国分离物外壳蛋白(Coat protein,CP)基因进行了扩增、克隆和测序,测序结果显示CP基因全长为1191nt,编码397个氨基酸。中国分离物与GenBank中其他分离物核苷酸序列同源性为70.2%~91.7%。系统进化分析显示不同ASPV分离物分为3个大的类群,呈现一定的寄主相关性,并未呈现明显的地理相关性。第一和第三大类群的寄主皆为苹果,第二大类群包含6个梨分离物和2个苹果分离物。不同ASPV分离物的抗原指数差异较大,推断可能由于序列变异引起抗原表位变异,从而引起血清型差异。
