阳离子交换树脂固定化乳糖酶的研究

摘    要:目的]以阳离子交换树脂为载体,研究黑曲霉来源乳糖酶固定化条件,以期改善乳糖酶性质,使黑曲霉乳糖酶更适于低乳糖乳的连续生产。方法]以吸附率最高的阳离子交换树脂D151为载体,通过先吸附后交联的方法固定乳糖酶,优化固定化条件。结果]结果表明,加酶量为50.0U/g(载体),吸附pH值为4.0,吸附温度是25℃,吸附时间24h,交联剂戊二醛浓度为4%,交联温度是30℃,交联时间是6h,固定化效果最好。获得的固定化酶活力可达11.8U/g(载体),固定酶回收率为37.2%。结论]该研究为工业化利用固定化乳糖酶连续生产低乳糖乳提供了技术依据。
