摘    要:利用无核白和黑王等12个品种的无病毒葡萄试管苗叶片为外植体,在NN69培养基(Nitsch and Nitsch 1969)试验了不同浓度的IBA和BA对其植株再生的影响.其中无核白、黑王、红地球和红宝石4个品种获得了稳定高频率的再生植株.以GUS基因为报告基因,通过根癌农杆菌介导,建立了葡萄离体叶片外源基因的遗传转化体系,而后将葡萄扇叶病毒外壳蛋白(GFLV-CP)基因转入葡萄叶片,通过卡那霉素筛选和PCR检测证明获得了转葡萄扇叶病毒外壳蛋白(GFLV-CP)基因的无核白葡萄植株.

Abstract:On the basis of high and stable plant regeneration from the leaves of "Tomson Seedless","Heiwang","Red Globe" and "Ruby Seedless" on the medium of Nitsch and Nitsch(1969),the transformation system of grape was established with GUS gene via Agrobacterium tumefaciens.The transgenic plant of "Tomson Seedless" was obtained with GFLV-CP gene by the selection of kanamycin and Conformed by PCR detection.
